Objective:Observe the effect of Huatan Tongyu Jiedu Decoction on epithelial-mesenchymal transition,invasion and metastasis of gastric cancer MKN-45 cells.Methods:1.Transforming growth factor-?1(TGF-?1)induced human gastric cancer MKN-45 cell epithelial-mesenchymal transition model;2.Observe the effect of different concentrations of Qitongtongjiedufangfang water extract on the invasion and metastasis of human gastric cancer MKN-45 cells induced by TGF-?1;observe the migration ability of human gastric cancer cells by cell scratch method and Transwell chamber method,respectively.Impact of invasive ability;3.To observe the effects of water extracts from different concentrations of Qitongtongjiedufang on TGF-?1-induced epithelial-mesenchymal transition-related markers in human gastric cancer MKN-45 cells;Western blot was used to detect human gastric cancer MKN-45 cells E-cadherin The expression of N-cadherin,Vimentin and Snail proteins was detected.The expression of E-cadherin,N-cadherin,Vimentin and MMP-7,9 proteins in human gastric cancer MKN-45 cells was detected by ELISA.1.Scratch test results showed that compared with the blank group,the wound healing rate of the model group significantly increased(P <0.01);compared with the model group,Huayu Tongqiao Jiedu Fang 10?g/mL,20?g/mL,40?g/ The healing rate of scratches in the mL group was significantly reduced(P<0.05 or P<0.01),and the healing rate of the scratches was dose-dependent(P<0.05 or P<0.05)in each dose group(5-40)?g/mL.0.01);Transwell invasion assay results showed that compared with the blank control group,the number of cells in the model group significantly increased(P<0.01);compared with the model group,Huayu Tongqiao Jiedufang 10?g/mL,20?g/mL,40?g/mL The number of cells in the dose group was significantly reduced(P<0.01),and it was dose-dependent in the(10-40)?g/mL group(P<0.05 or P<0.01).Western blot analysis showed that compared with blank group,the expression of N-cadherin,Vimentin and Snail protein in the model group was significantly up-regulated(P<0.01),and the expression of E-cadherin protein was significantly down-regulated(P<0.01);In comparison,the expression of N-cadherin,Vimentin and Snail protein was significantly down-regulated(P<0.05 or P<0.01)and the expression of E-cadherin was significantly up-regulated in Huayu Tongbi Jiedufang 10,20,and 40?g/mL dose groups.P<0.05 or P<0.01);a dose-dependent manner between(5-40)?g/mL dose groups;4.ELISA results showed that compared with the blank group,the content of N-cadherin,Vimentin and MMP-7,9 in the gastric cancer MKN-45 cells in the model group was significantly increased(P<0.01),and the content of E-cadherin was significantly down-regulated(P< 0.01);Compared with the model group,the contents of N-cadherin,Vimentin,and MMP-7,9 in the 10,20,and 40 ?g/mL groups of Huayu Tongbi Jiedufang were significantly down-regulated(P<0.05 or P<0.01).),E-cadherin content was significantly upregulated(P <0.05 or P <0.01);in a dose-dependent manner between(5 ~ 40)?g / mL dose group.Conclusion:1.Huatan Tongyu Jiedu Decoction can inhibit the invasion and migration of human gastric cancer cell MKN-45.2.Huatan Tongyu Jiedu Decoction can inhibit the invasion and metastasis by regulating the markers of MKN-45 epithelial-mesenchymal transition in human gastric cancer cells.
