BackgroundBarrett's esophagus(BE)is the most serious complication of gastroesophageal reflux disease(GERD)and the most important precancerous lesion of esophageal adenocarcinoma.In western countries,the incidence of BE is increasing year by year.Currently,the mechanism of occurrence of BE has not been elucidated.In the past,it was thought that long-term acid exposure can cause BE.However,some scholars had suggested that the role of bile acids is even more important in the development of BE.ObjectiveThis experiment was to study the effect of different concentrations of bile acids on the morphology and ultrastructure of BAR-T cells,and to explored the mechanism and clinical significance of BE metaplasia.Methods(1)Human Primary Esophagus BAR-T Cells,and then in the center laboratory of our hospital,and carried out passaging,cryopreservation,resuscitation and chemical stimulation.(2)Stimulated experimental subjects 3 times a day with 250umol/L,400umol/L,and500umol/L bile acid for 7 days.The morphological changes of the cells were observed under an inverted phase contrast microscope,and ultrastructural changes of the cells were observed under a transmission electron microscope.Results(1)The normal morphology of the BAR-T cells were found under the microscope in thecontrol group of cells.When cells grew to 7 days,contacting inhibition and apoptosis occured to most of them.In the experimental group,the growth time of cell was longer than that of the control group,normal contact inhibition and apoptosis in the experimental group was found rare under the microscope.(2)Under transmission electron microscope: the normal cell microstructure could be seen in the control group.The cells of experimental group showed mitochondrial swelling and deformation,nucleolar edge collection,dinucleotide and autophagy body formation.Conclusion(1)Stimulated by bile acids,the morphology and structure of BAR-T cells were changed.(2)Different concentrations of bile acids acted differently on BE cell lines.With the increase of the concentration of bile acid,BAR-T cells had the tendency of dysplasia.
