Part 1 Methylation changes of RASSF1 A and APC genes in NSCLCObjective: To investigate the methylation changes of RASSF1 A and APC genes after the effect on the growth,proliferation,invasion and apoptosis of lung cancer A549 and H358 cell lines when treated with 5-azacytidine.Methods : First,A549 and H358 cell lines were treated with 5-azacytidine(3?mol/L)for 24 h and 48 h.Cell cycle distribution was detected by flow cytometry,and cell invasion was measured by Transwell method.Hearst staining was observed under fluorescence microscope for apoptosis.Second,m RNA and protein expression of RASSF1 A and APC genes were detected by RT-PCR and Western blot in two cell lines after 5-azacytidine treatment.Results: After treatment with 5-azacytidine,the growth of A549 and H358 cell lines gradually stagnated,the invasive ability was weakened,the apoptosis increased.There was a positive relationship between invasive ability and degree of apoptosis and effect time;The transcription and expression of RASSF1 A and APC genes were increased in the two groups of lung cancer cell lines.Conclusions: 5-Azacytidine can weaken or even lose the function of lung cancer cells through gene demethylation.In this process,demethylation of promoter regions of RASSF1 A and APC genes are involved.Part 2 Methylation of NSCLC-related tumor suppressor genes by METAObjective: DNA methylation has been identified as a potentially powerful tool for tumor detection and diagnosis.However,the possibility of using methylation of tumor suppressor genes in blood as a biomarker for NSCLC detection and screening remains to be elucidated.This meta-analysis is designed to evaluate the value of methylation of tumor suppressor genes detected in peripheral blood samples as a non-invasive biomarker for the diagnosis of NSCLC.Methods: The CNKI database,Wanfang database,Chinese Sci-tech periodical database,PUBMED,and EMBASE were searched by computer and there was no time limit for retrieval.The retrieved documents were screened and 27 articles were eventually selected for inclusion in the study.Results: Twenty-nine NSCLC-related tumor suppressor gene methylation were screened out,of which P16,RASSF1 A,APC,RAR?,DAPK,CDH13,and RUNX3 gene promoter methylation had significant correlation with NSCLC.There was no significant correlation between sex and smoking,age,gender,tumor stage,and histology.Conclusions: The methylation status of peripheral blood tumor suppressor genes P16,RASSF1 A,APC,RAR?,DAPK,CDH13,and RUNX3 gene promoters can be used as a biochemical marker for early screening for NSCLC.Its clinical popularity requires further large-scale,large-sample studies to confirm. |
