The Expression And Function Of Long Non-coding RNA In Hepatocellular Carcinoma

Objectives: This study aimed to investigate the expression of long non-coding RNA MT1 JP in Hepatocellular carcinoma tissues and normal tissues.And we explore the relationship between the relative expression level and the clinical information,and the effect on the proliferation,invasion,migration,cell cycle and apoptosis of hepatoma cells.Methods: Quantitative reverse-transcription PCR(qRT-PCR)was performed to detect expression of MT1 JP in Hepatocellular Carcinoma(HCC)tissues and paracancer tissues to explore the differences of the clinical indicators of HCC between two groups of low and high expression.We constructed of lncRNA MT1 JP overexpression plasmid,transfected it into liver cancer cells with a series of cell phenotype experiments,including transwell experiments and flow cytometry,and observed the effect of lncRNA MT1 JP on liver cancer cell function.Results: The lower expression of MT1 JP was found in HCC tissue compared with the paracancer tissues(0.0018 vs 0.0301,p<0.05).According to the expression,Patients were divided into two groups.Its differences of theascites ? splenomegaly organ violation and serum Carcinoembryonic antigen were statistically(p<0.1).Overexpression of MT1 JP,liver cancer cell invasion and migration were inhibited(p <0.05).Conclusion: Long non-coding RNA MT1 JP is markedly downregulated in HCC tissue and has significant correlation with ascites,splenomegaly organ violation and serum Carcinoembryonic antigen.And the overexpression of MT1 JP can inhibit the invasion and migration of hepatocellular carcinoma cells,suggesting that lncRNA MT1 JP may be a new tumor suppressor gene of liver cancer.
