Background Lung cancer is the most common malignancy with the highest incidence and mortality in the world.Research on it has been a hot topic.With the continuous development and innovation of medical technology,the survival rate of lung cancer has been greatly improved,but the pathogenesis of lung cancer remains unclear.Therefore,it is urgently needed to explore the molecular mechanisms of the occurrence and progression of lung cancer and to seek specific high-level diagnostic and therapeutic targets.Long non-coding RNA(lncRNA)LINC00968 as a new lncRNA,its mechanism of action in lung cancer is still unclear.In this experiment,LINC00968 was used for bioinformatics prediction and analysis,combined with chip detection,cell transfection techniques,etc.to explore the regulatory mechanism of LINC00968 in lung adenocarcinoma(LUAD),with a view to providing treatment for lung adenocarcinoma The new therapeutic target.Methods In order to explore the expression and mechanism of LINC00968 in lung adenocarcinoma(LUAD),q RT-PCR was used to detect the expression of lncRNA in 20 clinical LUAD tissues,paracancerous tissues and lung cancer A549 cells.Low expression in LUAD tissues and A549 cells.Furthermore,we overexpressed lncRNAs in A549 cells and detected them with miRNAmicroarrays.We predicted and analyzed the possible target genes of these miRNAs and obtained a preliminary control of lncRNA-miRNA-mRNA.Network,and further verified by TCGA(The Cancer Genome Atlas)database,to explore the possible mechanism of LINC00968 in LUAD.Results 1.LINC00968 lung adenocarcinoma is lowly expressed in clinical specimens,which is consistent with the data in the TCGA database,suggesting that it has the potential for tumor suppressor genes.2.Combining the mi RNA chip detection technology and bioinformatics methods,we can see that the five miRNAs with significant differential expression are closely related to the pathogenesis of cancer.3.A preliminarily established regulatory network for LINC00968 was used to validate the network using data from patients with lung adenocarcinoma in the TCGA database and a validated lncRNA-mi RNA-mRNA regulatory network was obtained.Conclusion 1.LINC00968 was significantly low expressed in lung adenocarcinoma tissue and had the function of tumor suppressor gene,which could inhibit the development of cancer to a certain extent.2.LINC00968 can regulate the development and prognosis of lung adenocarcinoma through lncRNA-mi RNA-mRNA,and initially reveals its possible mechanism of action.3.The potential of LINC00968 tumor suppressor is expected to continue to explore as a therapeutic target for lung adenocarcinoma.
