Cancer immunotherapy is attracting more attention as a next generation cancer therapeutic strategy due to its ability to harness the innate immune system to attack tumor cells.Without coordinated strategies to mitigate the immunosuppressive nature of the tumor microenvironment,cancer immunotherapy generally offers limited clini-cal benefit for established tumors.Tumor associated macrophages(TAMs)are the critical driver of this immunosuppressive tumor microenvironment,which not only suppress T-cell activation but also promote tumor metastatic dissemination and impart resistance to cytotoxic therapies.The reprogrammed TAMs orchestrate cytotoxic lym-phocyte(CTL)recruitment in the tumor and direct memory T cells and B cells toward tumoricidal responses.This strategy could efficiently eradicate tumors,inhibit metas-tasis,and further prevent their recurrence,which holds tremendous promise to realize a potent cancer immunotherapy.
