| Objective:To investigate the effect of Brucea Javanica oil emulsion(BJOE)on the expression of scar fibroblasts TGF-beta 1,Smad2 protein and Smad2 mRNA,so that we can preliminary understanding if there is a potential therapeutic effect of BJOE on scar.Methods:The experiment was divided into 2 groups,experimental group:scar fibroblasts+DMEM+BJOE,control group:scar fibroblasts+DMEM,and the two groups of cells were intervened with 24h,48h,72h respectively,the expression of TGF-β1、Smad2 protein and Smad2 mRNA were detected by WB and RT-PCR respectively.Results:Compared with the control group,the expression of TGF-β1,Smad2 protein and Smad2 mRNA was significantly reduced in experimental group,and the difference between the two groups was statistically significant(P<0.05).With the prolongation of drug intervention time(24h-48h-72h),the expression of.Smad2 protein and Smad2 mRNA showed a decreasing trend,which was time-dependent.Conclusion:BJOE can reduce the expression of TGF-β1,Smad2 and Smad2 mRNA in scar fibroblasts. |
