Obective: Xanthium has the function of dispelling wind,relieving pain through orifices,and relieving pain.It is used as an important medicine for the treatment of nasosinusitis in traditional Chinese medicine.The application of Fructus Xanthium has a long history and the curative effect is remarkable,but its active components and mechanism of anti-allergy are not clear at present.The mechanism of the treatment of allergic rhinitis was preliminarily discussed in order to provide a safe and reliable scientific basis for the clinical application of Fructus Xanthii..Methods: In animal experiments,Establishment of allergic rhinitis model in mice combined ovalbumin ovalbumin(ovalbumin)intraperitoneal injection with nasal drip stimulation,and divided into different groups..The mice were treated with water extract of Fructus Xanthium,and the behavioral symptoms of nasal scratching and sneezing were observed.After the administration,we take blood,separate serum.and the nasal lavage fluid,nasal tissue and spleen were obtained for pathological examination.In cell experiment,Fructus Xanthii was co-cultured with human myeloma cells(U266 cells).When the experiment is over,the levels of IgE,IL-5 and cytokines in culture supernatant were determined by ELISA.Results: The observation and statistics of animal behavior showed that,Compared with rhinallergosis model group,the nasal scratching and sneezing number of mice treated with medium and high dose of Xanthium were significantly decreased,the results of pathological sections showed that,compared with the rhinallergosis model group,the nasal mucosal epithelial cell damage was reduced,tissue edema was decreased,nasal congestion was alleviated,eosinophilic granulocyte infiltration was significantly decreased in the middle and high dose group of Xanthium sibiricum,Pathological section of spleen can be seen that Compared with the allergic rhinitis model group,the white pulp of the high-dose group of Xanthium sibiricum increased obviously,the lymphocyte count increased,and the pathological symptoms were improved significantly;The results of ELISA assay showed that the extract of Fructus Xanthii could significantly reduce the level of IgE secreted by U266 cells,meanwhile,the content of IL-5 in the cell culture fluid decreased with the increase ofthe dose of the extract of Xanthium cubeba by the cell experiment.Conclusions: The results of animal experiments showed that the group treated with medium and high dose of Xanthium could significantly improve the typical symptoms of allergic rhinitis in mice,reduce sneezings and nose scratches and at the same time,nasal mucosa histomorphology showed that high dose of Xanthium could significantly relieve epithelial cell injury,improve tissue edema,reduce congestion of nasal mucosa and decrease eosinophil infiltration,the pathological section of spleen showed that,the white pulp and the count of Lumbar fine cells in the high dose group of Cangezi were significantly larger than those in the former group,and the count of Lumbar fine cells was increased in the high dose group.the results suggest that Xanthium has a certain the rapeutic effect on allergic rhinitis in mice;The results of ELISA assay showed that the extract of Fructus Xanthium could significantly reduce the level of IgE secreted by U266 cells,meanwhile,the content of IL-5 in the cell culture fluid decreased with the increase of extract dose of Xanthium cubeba,the results suggest that the extract of Fructus Xanthium may reduce the production of IgE by inhibiting the secretion of IL-5.
