Phellodendron amurense Rupr.(Rutaceae)is a deciduous tree commonly called the Amur cork tree.The bark of P.amurense has been widely used as a traditional medicine in China.P.amurense produces Atrorrhizine,Palmatine and Berberine.These alkaloids were regarded as rich source of pharmaceutically important biologically-active components.This study investigate of supercritical C02 fluid extraction of alkaloids from natural phellodendron amurense process conditions,and discusses the influence on the temperature,pressure,time,ethanol volume fraction of phellodendron amurense and alkaloid content of three kinds of alkaloids and the total alkaloids.By using the response surface method optimization of supercritical CO2 fluid extraction of alkaloids from natural phellodendron amurense process conditions.(1)This study investigate of the single factor experiment on supercritical CO2 fluid extraction of Atrorrhizine,Palmatine,Berberine and the total alkaloids from natural phellodendron amurense,and discusses the factors about temperature,pressure,time and ethanol volume fraction.And determine selected 30Mpa to be the extraction pressure,50? for the extraction temperature,60min for the extractions time and 95%of ethanol for the extraction of entrainer.(2)To extract the Atrorrhizine,Palmatine,Berberine and total alkaloid from Phellodendron amurense Rupr using supercritical CO2 extraction and to optimize the extraction conditions,the effects of supercritical C0O2xtraction pressure,temperature,time and the volume of ethanol on extraction the total alkaloid were studied.Base on the single factor test,a Box-Behnken design involving four factors at three levels was generated by the Design-Expert software and the response surface methodology was used to optimize supercritical CO2 extraction conditions.The optimized conditions were as follows:30Mpa of the extraction pressure was,50? of extraction temperature was,90 min of extracting time was,and 95%of the volume of ethanol.The actual Atrorrhizine,Palmatine,Berberine and total alkaloid from Phellodendron amurense Rupr was 0.987mg/g,0.066mg/g,0.787mg/g and 1.79 mg/g closed to the predicted yield.The supercritical CO2 extraction conditions of from Phellodendron amurense Rupr using Box-Behnken design and response surface methodology was well established and optimized.(3)This experiment use five different oxidation methods to comprehensive evaluation of the capacity in antioxidant about different extraction method from phellodendron amurense.In the range of the concentration about the test.The extraction of substances have strong clearing ability on DPPH,O2-andOH the and to resist lipid peroxidation,and restore the ability of Fe3+ in the a phellodendron amurense from the supercritical CO2 extraction,ultrasonic extraction and soxhlet extraction.Their antioxidant capacity is increased with the change of the concentration,reaches maximum value when the concentration is the range of the test.Explain phellodendron amurense extracted by three extraction methods of material had well antioxidant activity,and the size of the antioxidant capacity is the supercritical CO2 extraction>ultrasonic extraction>soxhlet extraction.
