Objective: The purpose of this study was to investigate the expression of YAP and Mdm2 in invasive breast cancer of different molecular subtypes. The biological characteristics of YAP and Mdm2 were analyzed in different molecular subtypes of breast cancer, their development, invasion and Metastasis,to study whether they can become a prognostic factor to determine the prognosis of different molecular subtypes of breast cancer, but also for the Basal-like breast cancer targeted therapy to provide ideas and basis. Methods: The sample was taken from tissue samples of 124 breast cancer patients confirmed by pathology between January 2015 and January 2017 in the First Affiliated Hospital of Nanhua University.Immunohistochemical PV two-step method was used to detect the expression of yap and mdm2 protein in different molecular subtypes of breast cancer and to analyze its relationship with the clinicopathological features of breast cancer patients. The results of this study using SPSS21.0 statistical software for analysis, using X2 test or Fisher exact test for significance, take P <0.05 was considered statistically significant.Result:1.There was a statistically significant difference in the expression of Yap in breast cancer tissues of different molecular subtypes(P=0.029).Compared with each other,Luminal B was significantly higher than Basal-like and Her-2 overexpression was significantly higher than Basal-like and Luminal A,with statistically significant differences(P<0.05).Comparison of Luminal B and Luminal A,Luminal B and Her-2 overexpression, and Luminal A and Basal-like showed no significant differences(P>0.05).2.There was a statistically significant difference in the expression of Mdm2 in different molecular subtypes of breast cancer(P=0.012).Compared with each other,Luminal A was significantly higher than Her-2 overexpression and Basal-like was significantly higher than Her-2 overexpression, with a statistically significant difference(P < 0.05).Comparison of Luminal B and Her-2 overexpression,Luminal A and Luminal B,Luminal A and Basal-like and Luminal B and Basal-like showed no significant differences(P>0.05).3.The positive expression of Yap in 124 breast invasive ductal carcinomas was not significantly associated with menstrual status, histological grade, lymph node metastasis,TNM staging,ER/PR status, and Ki-67 index(P>0.05).It was significantly correlated with age,Her-2 expression and tumor size(P<0.05).The positive expression rate in the group older than 50 years was higher than that in the age group <50,the positive rate in the Her-2 positive group was higher than that in the Her-2 negative group, and in the tumor size <2cm group and 2cm-5cm group positive expression rate is higher than >5cm group.4.The positive expression of Mdm2 in 124 breast invasive ductal carcinomas was not significantly related to menses status, lymph node metastasis, tumor size, histological grade,TNM staging,Ki-67 index, and Her-2 expression(P>0.05).There was a significant correlation with age and ER/PR status(P<0.05).The positive expression rate was higher in the age group <50 years than in the age group ? 50 years, the positive rate in the ER/PR expression ? 1% group was high than in ER/PR expression <1% group.Conclusion:1.Yap has significant differences in the expression of different molecular subtypes of breast cancer, and has a significant correlation with Her-2 expression. It has important significance for the diagnosis and treatment of Her-2 over-expression breast cancer..2.Mdm2 expression is significantly different in different molecular subtypes of breast cancer, and has certain reference value for the diagnosis and treatment of Luminal A and Basal-like breast cancer.
