Background: In our research group has conducted a lot of research on the pharmacodynamics and mechanism of Xiaoyao Powder in antidepression.However,Xiaoyao Powder is composed of 8 herbs,the scientific connotation of its compatibility is not yet clear.In particular,the compatibility of chemical components is complex and the law of change is less clear.Explaining the connotation of Chinese herbal compound is an important task of modern Chinese medicine research.As the basic unit and the most common form of Chinese traditional medicine compatibility,it can reveal the law of compound compatibility.Therefore,taking drug pair as the breakthrough point,it is necessary to explore the law of compound compatibility.It can reveal the theoretical basis of prescription compatibility and provide reference and basis for exploring the law of compound compatibility.The compatibility drug pairs in Xiaoyao Powder include Bupleuri Radix-Paeoniae Radix Alba,Bupleuri Radix-Menthae Haplocalycis Herba,Angelicae Sinensis-Radix-Paeoniae Radix Alba and Atractylodis Macrocephalae Rhizoma-Poria.Bupleuri Radix-Paeoniae Radix Alba,as the most basic drug for dispersing the liver and nourishing the liver,are the core drugs for many antidepressant compounds such as Xiaoyao Powder,Chaihu Shugan Powder,Sini San,etc.Bupleuri Radix soothes liver and dissolves stagnation,Paeoniae Radix Alba nourishes soft liver.The two drugs are used together to promote each other,interdependence,supplementing liver blood.At present,researchers have also done some research on the changes of chemical composition after the combination of Bupleuri Radix-Paeoniae Radix Alba.It was found that the amount of paeoniflorin could be increased and the amount of benzoic acid could be decreased in Paeoniae Radix Alba after the two decoction compatibility.But the change of the total chemical composition was not clear.The change of chemical composition before and after the compatibility of Bupleuri Radix-Paeoniae Radix Alba is the material basis of its compatibility and synergism,and the chemical mechanism of its compatibility is the starting point to study the pharmacological mechanism of the compatibility.Therefore,this paper focuses on the study of the chemical mechanism of the compatibility of Bupleuri Radix-Paeoniae Radix Alba and its contribution to Xiaoyao Powder.Objective:(1)The changes of chemical constituents before and after compatibility of Bupleuri Radix and Paeoniae Radix Alba were analyzed by background subtraction of mass spectrometry combined with metabolic technique,and the chemical mechanism of synergistic effect between them was revealed.(2)The chemical compositions of Bupleuri Radix and Paeoniae Radix Alba and Xiaoyao Powder were compared and analyzed,and the modern scientific connotation of the drug on the core status in compound antidepressant effect was stated.Methods:(1)The chemical constituents of the water extract and alcohol extract of Bupleuri Radix,Paeoniae Radix Alba were analyzed by UPLC-MS/ MS method.(2)By using the method of plant metabonomics and background subtraction(BSA)in mass spectrometry,we can comprehensively analyze the chemical constituents with different contents of ethanol extract from Bupleuri Radix(Paeoniae Radix Alba).(3)The chemical compositions of water extract of Xiaoyao power were analyzed by UPLC-MS/MS method,the chemical components of Bupleuri Radix and Paeoniae Radix Alba were assigned to the chemical composition of Xiaoyao Powder.Results:(1)The chemical constituents of water extracts and alcohol extracts from Bupleuri Radix and Paeoniae Radix Alba were analyzed by UPLC-ESI-Q/exactive orbitrap technique.Finally 70 peaks were detected in the water extract of Bupleuri Radix,31 compounds were identified;42 peaks were detected in the ethanol extract of Bupleuri Radix,and 14 compounds were identified.A total of 60 peaks were detected in the water extract of Paeoniae Radix Alba,43 compounds were identified.34 peaks were detected in the ethanol extract of Paeoniae Radix Alba,and 20 compounds were identified.(2)The overall difference of chemical components in vitro before and after compatibility of Bupleuri Radix and Paeoniae Radix Alba was analyzed by using metabonomics combined with UPLC-MS/MS background substraction method.The results showed that the components of peaks 14,peaks 20,peaks 21,and 56 in Bupleuri Radix,and paeoniflorin,galloyl paeoniflorin or its isomers,peaks 15,peaks 16 in Paeoniae Radix Alba were increased after the compatibility.The components of saikosaponin a,saikosaponin c,peak 41,2''-O-acetylated saikosaponin a,3''-O-acetylated saikosaponin a,4''-O-acetylated saikosaponin a,peak 53,peak 54,peak 55,peak 57 in Bupleuri Radix,peak 10 in Paeoniae Radix Alba were decreased after the compatibility.(3)The chemical constituents of Bupleuri Radix and Paeoniae Radix Alba and Xiaoyao Powder were assigned by UPLC-ESI-Q/exactive orbitrap technique.In positive ion mode,there were 77 chromatographic peaks in Xiaoyao Powder were detected,and 30 peaks were from Paeoniae Radix Alba,27 from Bupleuri Radix.In negative ion mode,there were 86 chromatographic peaks in Xiaoyao Powder were detected,35 were from Paeoniae Radix Alba and 18 from Bupleuri Radix.Conclusion:(1)In this paper,the method of metabolic technique combined with background subtraction of UPLC-MS/MS mass spectrometry was established to analyze the changes of chemical composition of Bupleuri Radix and Paeoniae Radix Alba before and after compatibility,which laid a foundation for elucidating the compatibility mechanism of Bupleuri Radix and Paeoniae Radix Alba antidepressants.(2)The contribution of core drugs in Xiaoyao Powder to the compound of Bupleuri Radix and Paeoniae Radix Alba was analyzed by UPLC-MS/MS,which laid a foundation for elucidating the basis of antidepressant substance and compatibility of Xiaoyao Powder.
