Spinal cord injury(SCI)has a high incidence of clinical symptoms.The series of clinical symptoms caused by spinal cord injury seriously affect the quality of life of patients,and no effective treatment method has been found.The research results show that neural stem cells(neural stem cells,NSCs)highly self-renewal and pluripotent differentiation,proliferation and migration and so on many characteristics,use of NSCs for spinal cord injury,can be a function cells,directional differentiation in lesions in repair damaged spinal cord tissue plays an important role.This experiment mainly study simple neural stem cells,after local injection transplantation and subarachnoid injection transplantation,5 levels deep analgesia SCI model the influence of repair,and neural stem cells to treat SCI clinical research work to lay a solid foundation.In this study,16 healthy beagle dogs aged 18 months were selected randomly into 4groups.Model group respectively,fresh and local injection treatment group,old gen NSCs NSCs subarachnoid injection treatment group and fresh gen NSCs subarachnoid injection treatment group,and in the back of the dog on the left side of the L2 section of spinal cord with balloon compression method,5 levels deep analgesia SCI model is set up.1 d before making model,made the mold after 1 d,3 d,7 d and 14 d and 21 d and 28 d,44 d,56 d for each group of experimental canine behavior score and blood,and measure its physiological indexes of blood,biochemical index propagated,CREA,ALP,TP,Ca,UA,LDH,AST,ALT,CRP and IL-10 anti-inflammatory agents and PGE2,TNF-a,anti-oxidation factor of SOD,MDA,T-AOC,nerve expression of Nestin factor,SHH.1d,3d,7d,14 d,28d and44 d after 1d and molds were performed,and each group of beagle dogs were randomly assigned to an mri scan for damage to the spinal cord structure.Test results:1.5 levels deep analgesia than the case into a dog SCI model of experimental research,found that oppression quantity control in 5 ml and oppression time control within 10 min,the beagle of 10 + 1 kg permanence paralysis of spinal cord injury model,can achieve common persistence paralysis of spinal cord injury clinical effect.2.Through local injection transplantation and spinal epidural injection of these twokinds of ways to transplant neural stem cells in the spinal cord injury the body,can reduce the number of mononuclear cells,reduce the spinal cord injury occurred secondary inflammatory reaction.3.Neural stem cell transplantation can reduce the phenomenon of osteolysis in the body of spinal cord injury,participate in the regulation of blood calcium,and play a protective role in renal function.4.Whether it's old gen NSCs subarachnoid injection treatment group,or fresh gen NSCs subarachnoid injection treatment group compared with local injection group,in spinal cord injury after subarachnoid injection can improve total beagle better anti-inflammatory ability,protect spinal cord injury tissue,prevent excessive oxidation,prevent issued after spinal cord injury.5.In the early stage of acute and chronic spinal cord injury,neural stem cells can significantly improve SHH content and help repair the spinal cord injury.In the late stage of repair of spinal cord injury,the body can produce endogenous SHH without improving any treatment,which can help to repair the injury.6.NSCs can be used to treat spinal cord injury by subarachnoid injection,which can rapidly increase the concentration of prostaglandin E2 in dogs with spinal injury.7.It is indicated that the activity of lactate dehydrogenase in the blood can be improved after spinal cord injury for a longer time after spinal cord injury.Local injection of NSCs can regulate the activity of lactic dehydrogenase in earlier time,but a short time,and subarachnoid injection of NSCs of lactate dehydrogenase activity regulation no local injection transplantation early,but to maintain for a long time,is advantageous to the repair of spinal cord injury.
