CBL-B Protein Can Treat The Expression Of EGFR In Non-small-cell Lung Carcinoma By Inhibiting Activation Of EGFR

Objective: To illuminate the role of CBL-B protein in the treatment of EGFR-expression in Non-small-cell lung carcinoma(NSCLC)by inhibiting the phosphorylation of EGFR.Methods: A549 cell is a kind of NSCLC expressing EGFR?The stable over expression of CBL-B A549 cell line(named CB-A549)was fabricated by CBL-B lentiviral activation particles.A549 cell lines were transfected by viral vectors,the A549 cell lines were stabilized after 72 hours and the western blotting was used to confirm that the cell line could over express CBL-B protein after 5 generations.The A549 cell line,which can stabilize the over expression of CBL-B protein,is renamed as CB-A549.The CCK-8 test was performed to detect the proliferation ability of A549 and CB-A549 cells.The ability of proliferation of A549 and CB-A549 was detected by CCK-8 assay,the expression of EGFR in cell lines were detected by western blotting.Results: The ability of proliferation,migration of CB-A549 was attenuated compared with A549 cell line.Apoptosis of CB-A549 was increased obviously contrasting with A549 and activation of EGFR in CB-A549 was decreased compared with A549.Conclusion: CBL-B protein can with negatively regulating the activation of EGFR in NSCLC,CBL-B protein can decrease the ability of proliferation,cell migration,apoptosis of A549 cell.
