Objective At the early stage of the subject of this research study found gallbladder's heat method(on behalf of Biyuanshu oral liquid)work by inhibiting advantage bacterial biofilm formation of staphylococcus in nasal cavity.Detect the expression of biofilm forming-associated agr,luxS,atlE gene system by rt-pcr technique,in order to explore the complex regulatory mechanism of inhibiting BF from the gene regulation.Methodology A total of 8 patients with chronic rhino-sinusitis of stagnated heat of gallbladder symptom enrolled from Sichuan-yonghcuan region were classified into treatment group.2 healthy people with no treatment were classified into control group.The treatment group received Biyuanshu oral liquid produced by Huashen company for 14 days.We took the pre-treatment and post-treatment secretion of nasal cavity on the same side as samples,which dominant bacteria is staphylococcus.Investigated the pre-treatment and post-treatment biofilm forming.Among them,8 cases were included as experimental group which accorded with asal epidermis staphylococcus aureus biofilm(+)before and after treatment to the nasal epidermis staphylococcus aureus biofilm(-).The samples before treatment were experimental group 1,after treatment were experimental group 2,and 4 cases of healthy people were control group.Design and synthesis primers,using RT-PCR technique to detect the bacterial biofilm control gene: staphylococcus : agrA,luxS,atlE expression quantity before and after taking medicine,BB170 hartz vibrio luminescence method to detect the AI-2 molecular signal intensity,finally come to the conclusion by statistical analysis.Result1.Statistical results of all cases : The level of luxS was suppressed after treatment(P < 0.05),but it`s no difference with the health group in both per-treatment and post-treatment(P > 0.05).To the level of agrA gene and atlE gene,There was no difference not only between per-treatment and post-treatment,but also between the health group and both of them(P < 0.05).2.The statistical result of cases working through luxS/AI-2 pathways : The level of luxS was suppressed after treatment(P < 0.05),and it is lower than the healthy group in per-treatment(P < 0.05),while it`s no difference with the health group in post-treatment(P > 0.05).To the level of agrA gene and atlE gene,There was no difference not only between per-treatment and post-treatment,but also between the health group and both of them(P>0.05).3.The statistical result of cases working through luxS/AI-2 pathways : The level of agrA was reduced after treatment(P < 0.05),and it`s no difference with the health group in per-treatment(P > 0.05),but lower than the health group after treatment(P < 0.05).To the level of luxS gene and atlE gene,There was no difference not only between per-treatment and post-treatment,but also between the health group and both of them(P > 0.05).4.The correlation test results for the differences between different indicators showed that there was no correlation between the two indicators.Analysis of the linear correlation trends of the difference between agrA and atlE lines observed negative correlation trends.Conclusion1.In the nose deep sigh of oral liquid in the treatment of CRS mainly by inhibiting the advantage bacterium group bacteria biofilm work table Portugal,its membrane inhibition may work through complex film-forming gene regulatory network,a complex system may include luxS QS system key genes and related biological signal AI-2,agr system key genes agrA and related adhesion control gene atlE.2.The adjustment performance of the nasofushu oral solution to the key gene luxS was up-regulated,and by up-regulating the luxS gene,the ai-2 signal was decreased,and the negative regulation reached the effect of film inhibition.The regulation of the key gene agrA was down-regulated,and it was regulated to the effect of membrane inhibition.3.To the agrA gene and its related adhesion atlE gene,the complexity of the regulating performance has both raised and lowered,suggests that nasal deep sigh of oral liquid is not agr-atlE pathways through a single linear adjustment influence the formation of table keeps BF,but may be directly regulating the expression of atlE,or other complex pathways exist at the same time influence the expression of atlE.
