Wedelia chinensis is one kind of herbaceous perennial belonged to Wedelia of Compositate,which entire plant is widely used as traditional Chinese herbal drug that can treat sore throat,diphtheria,pertussis,dysentery,hemorrhoids,abdominal pain,rheumatism,traumatic injury and other diseases,which indicate some bioactive compounds must contained in the plant.It has the functions of heat-clearing and detoxifying,protecting liver and gallbladder,cooling blood and stopping bleeding,and diminishing inflammation and pain.In China,Wedelia chinensis can be found in the wild frequently and is mainly distributed in coastal areas such as Guangdong,Fujian and Taiwan,hence it is a common medicinal plant contains a variety of bioactive components.It has been found that Wedelia chinensis has many pharmacological effects,such as antivirus activity,antitumor activity,anti-inflammatory activity,analgesic function,liver preservation function,insecticidal function,antibacterial activity,hypoglycemic action,inhibiting angiogenesis,scavenging free radical,protecting nervous system,promoting wound healing,and preventing human body from osteoporosis,respectively.This study focus on the antibacterial activity of Wedelia chinensis,we analyzed its main effective components as well as separated and purified the active compound from them.The compounds were isolated and purified by means of series column chromatographies and their molecular structures were determined through spectroscopic techniques.Specifically,Silica gel column chromatography,Sephadex LH-20 gel column chromatography,C18 reversed silica gel column chromatography,high performance liquid chromatography?HPLC?and other column chromatography techniques were used for separation and purification.The compounds were analyzed and identified by ESI-MS,1H-NMR,13C-NMR,DEPT and other spectroscopic methods.Staphylococcus aureus,Bacillus subtilis,Streptococcus pyogenes,Escherichia coli,Shigella flexneri and Aeromonas hydrophila were selected for screening antibacterial active drugs in antibacterial tests.The antimicrobial activity was measured with broad-spectrum antibiotic gentamicin as positive control.The antibacterial activity was estimated via antibacterial circle measurement and minimum inhibitory concentration?MIC?determination as well as minimum bactericidal concentration?MBC?determination.Compound 1 was isolated and purified from petroleum ether extract of Wedelia chinensis.Results showed that compound 1 possesses strong bacteriostasis and bactericidal effects and broad-spectrum antibacterial efficacy.Compound 1 has significant inhibitory effects on Staphylococcus aureus,Bacillus subtilis,pyogenic Streptococcus and Aeromonas hydrophila.Concretely,the MIC of compound 1 was small to the above 4 experimental bacteria,which were 2.0?g/mL,1.0?g/m L,0.5?g/m L,2.0?g/m L,while the MBC of compound 1 to Streptococcus pyogenes and Aeromonas hydrophila were 1?g/m L and 2?g/m L respectively.In conclusion,the above experimental results showed that compound 1 isolated from the extract of Wedelia chinensis perform significant antibacterial efficacy,for which the content of compound 1 is high and the antibacterial effect is strong.Consequently,compound 1 is considered to be the main antibacterial active component in Wedelia chinensis,and was identified as enantiomer-16-alkenes-19-acid according to spectral data.Effective antibacterial active ingredients proved to be contained in the petroleum ether extract of Wedelia chinensis,which provide some medicinal value for pharmaceutical research and suggest that bioactive compounds extracted from Wedelia chinensis may be potential antibacterial as substitution for antibiotics.
