ObjectiveLong non-coding RNA(Lnc RNA),as a type of RNA with a transcript longer than 200 nucleotides in length,Because it lacks a complete open reading framework,it does not have a protein coding function.Research has shown that LncRNA plays a key role in the occurrence,development,and metastasis of prostate carcinoma.In this study,the expression of LncRNA expression in prostate carcinoma patients was analyzed by The Cancer Genome Atlas(TCGA)?Our group first discovered that the FAM66 c was highly expressed in high-risk prostate cancer patients.It may be a potential oncogene.For this reason,we further studied it and explored its intrinsic pathogenic mechanism.Methods1.Grouping was according to the high expression and low expression of FAM66 c in clinical prostate cancer patients,the relationship between the expression level and clinicopathological features was analyzed by chi-square test and Wilcoxon non-parametric test combined with clinical data.2.RT-PCR was used to detect the expression of FAM66 c in normal prostate stromal cells and prostate cancer cells.3.Effect on proliferation and migration of prostate cancer were detected by MTT colony formation assay and wound healing assays after specific small interfering RNA(FAM66c siRNA)knock down the expression of FAM66c4.Bioinformatics analysis the potential targets and cell signaling pathways of FAM66 c.Results1.FAM66 c is closely related to the clinical pathology of patients: The average recurrence time of tumors in patients with high expression of FAM66 c was 30.91 months,and the average time of disease in low expression group was more than 180 months.Patients in the high-expression group had a significantly worse prognosis(P=0.026).In the comparison of overall survival time,The average overall survival time of FAM66 c high expression group was 81.1 months.The average overall survival time of the low expression group was more than 180 months.The overall survival time of patients with high expression of FAM66 c was significantly shorter and the prognosis was poor(P=0.0000355).And FAM66 c is closely related to clinical pathological staging,pathological Gleason score and biochemical recurrence index(P<0.05).2.The expression of FAM66 c was significantly higher in prostate cancer cell lines DU-145,22RV1,and PC-3M-IE8 than in prostate normal stromal cells WPMY-1.3.Knock down FAM66 c with siRNA significantly inhibited the proliferation and migration of prostate cancer cells.4.Bioinformatics analysis showed that FAM66 c may affect tumor proliferation through EGFR and BCL2.ConclusionWe analyzed the LncRNA expression profile of prostate cancer patients in TCGA and found for the first time that the high expression of the oncogene FAM66 c could affect the prognosis of prostate cancer patients.Through bioinformatics analysis,it was found that FAM66 c may participate in the occurrence and development of prostate cancer tumors through various channels.In addition,we also confirmed that down-regulating FAM66 c can inhibit the proliferation and migration of prostate cancer cells.In conclusion,this study describes the function and possible mechanism of action of FAM66 c as a novel oncogene in prostate cancer cells.It provides new ideas for the diagnosis,treatment and prognosis of prostate cancer.
