Objective:To explore the effects of advanced glycation end products(AGEs)on the proliferation,invasion and epithelial mesenchymal transition of human colon cancer cell line SW480 and the possible mechanisms.Methods:Human colon cancer cell line SW480 were divided into control group,AGEs(200?g/mL)group and AGEs(200?g/mL)+AG490(50?mol/L)group.After treatment,cell proliferation were tested by CCK8 kit,cell cycle and apoptosis rate were analyzed by flow cytometry.The expressions of EMT marker E-cadherin,N-cadherin,Vimentin and the JAK2/STAT3 pathway molecule STAT3,p-STAT3,JAK2 and p-JAK2 were tested by Western blot.Results:1?Compared with the control group,advanced glycation end products(AGEs)can significantly promote the proliferation of colon cancer SW480 cells,reduce the G1/S phase arrest,inhibit apoptosis,increase the invasion and migration ability in vitro.These differences were statistically significant(p<0.05)2.Compared with the AGEs group,the proliferation of colon cancer SW480 cells in the AGEs+AG490 group was decreased,the G1/S phase arrest was increased,the apoptosis was increased,and the invasion and migration ability was decreased,These differences were statistically significant(p<0.05).3.Western blot showed that AGEs increased the expressions of N-cadherin,Vimentin and the JAK2/STAT3 pathway molecule p-STAT3,p-JAK2 and down regulated the expression of E-cadherin.AG490 reversed AGEs induced E-cadherin down regulation and N-cadherin,Vimentin,p-STAT3,p-JAK2 up regulation.Conclusion:1.AGEs can reduce the G1/S arrest of colon cancer SW480 cells and promote the proliferation of colon cancer sw480 cells.2.AGEs can promote colon cancer sw480 cell invasion and migration in vitro.3.AGEs can inhibit the apoptosis of colon cancer sw480 cells.4.The above effects of AGEs were achieved through the JAK2/STAT3 signal pathway.
