Objective: To investigate the characteristics of RBCs producted by lipid whole blood and nomal blood during routine storage to providing some reference for clinical blood use.Methods: RBC units produced by 3-level lipaemic whole blood were stored and sampled on Day 0,7,14,21,28,and 35.Hematological and biochemical analysis were used to evaluate the quality of stored lipaemic RBC during routine storage,and the metabolites of RBC on Day 0,14,and 35 were used as indexes of metabolic profile analysis.Results: Hematological parameters of control and lipaemic groups MAP RBCs during the routine storage were changed slightly.Potassium and FHb concentration accumulated in supernatant of high-level and very-high-level lipaemic RBCs significantly.Metabolites associated with energy metabolism(Glucose,Pyruvic acid,and Adenine)and with redox metabolism(GSH,GSSG,and Niacinamide)were distinctly different between high-level lipaemic and normal RBCs.Conclusion: In response to high oxidation environment and oxidative stress,glucose catabolites are channeled through the PPP to fuel the generation of NADPH and maintain GSH redox poise.Oxidation environment and oxidative stress lead to the alteration of biochemical parameters of lipaemic RBC in the early phase of storage.
