Recently,hydrogels have been extensively studied for the application in biomaterials,tissue engineering,drug delivery.Polyethylene glycol has good biocompatibility,innocuity and low immunogenicity.It has good application potential.However,conventional hydrogels are usually difficult to control,which is one of the important factors that restrict the development of hydrogels.In this paper,novel polyethylene glycol?PEG?hydrogel had been prepared by the method of catalytic oxy-Michael reaction,and the application in drug release was studied.The effects of different reactant ratios and different alcohol modifications on the mechanical properties and drug release properties of hydrogels were discussed.1.The PEG gel was synthesized through the catalytic oxa-Michael reaction of EG6 and DVS.Glycerol was employed to form crosslinking networks and DMAP was employed as catalyst.The effects of alcohol modification on the mechanical properties of PEG hydrogels were discussed.Different proportion of hydrogels were used to explore the drug release properties.The results show that the PEG hydrogel has adjustable water absorption.Different alcohols can further modify the synthesized PEG hydrogels and their mechanical properties were tunable.Oleanolic acid was used as drug model,and its drug release experiment showed that the ratio of different reactant could regulate the drug release rate.2.Using DOX as a hydrophilic drug model and oleanolic acid as a hydrophobic drug model,the drug release of different alcohol modified PEG hydrogels through single drug loading and co loading was explored.The results showed that PEG hydrogel could encapsulate the hydrophilic and hydrophobic drugs well,and the co encapsulate and release of hydrophobic and hydrophilic drugs can be achieved.Different alcohol modified hydrogels could adjust the drug release rate.Using MCF-7 and MCF/MDR cell line,the anti-cancer effect had been explored.The result showed DOX and OA has synergistic effect,and the co-loaded PEG hydrogels has better antitumor activity.3.With cyclohexane as reverse phase solvent,Span-80 and Tween-80 as surface active agents,PEG microgels had been prepared by reverse phase emulsion polymerization,and the application of drug release were preliminarily explored.The results show that the microgels with different size can be obtained by changing the proportion and dosage of the surfactants,and they can be applied to drug release.In this paper,novel PEG hydrogel was prepared by the method of oxy-Michael reaction.It has adjustable mechanical properties and drug release rate,and can be applied into the co encapsulate and release of hydrophilic and hydrophobic drugs.
