Tumor microenvironment plays an important role in the process of tumor genesis,invasion and metastasis and so on,and it is also closely related to tumor resistance to drugs.Therefore,the study of tumor microenvironment is very meaningful to solve the problem of tumor treatment.Compared with the traditional tumor research methods,the tumor microenvironment simulation and research based on microfluidic chip in vitro has some advantages.First of all,microfluidic chip has the advantages of highly integration,could achieve the goal of the combination of different functional units,which is conducive to a series of important metabolites on-line detection in tumor microenvironment.Second,the flexibility of the chip design makes the application of a variety of biocompatible materials can be achieved,which is letting the tumor microenvironment simulation closer to the body's state in vivo.What is more,the material exchange of tumor cells is a dynamic process,so the research and simulation of tumor microenvironment in vitro should also meet the conditions,and microfluidic chip can guarantee the real-time exchange of nutrients and metabolites through external power.Based on the above advantages of microfluidic chip in tumor microenvironment simulation,this paper mainly studied the three-dimensional dynamic co-culture of tumor cells and endothelial cells on the chip platform,and used it as a simple tumor microenvironment simulation method to study on the anti-tumor effect of antioxidants.Firstly,in this paper,the development process and application field of microfluidic chip were summarized.The characteristics of tumor microenvironment and the existing research methods are reviewed;especially the work of tumor-related cell co-culture,showing the microfluidic chip has advantages and meanings in microenvironment research.Secondly,the lumen structure was produced by the soft mold method in the modified porous gelatin hydrogel,and the endothelial cells were attached to the cavity surface to form a vascular structure,and the glioma cells suspended in the co-culture gel and the chip was used to antioxidant effect study for tumor cells reactive oxygen.The utilization of this platform gives a new way to study the co-culture conditions of different cells,and can be used as a drug screening research methods in vitro.Finally,we used the agarose hydrogel modified by RGD polypeptide as the cell spreading material and combined with the concentration gradient simulation.The characteristics of the co-culture of glioma cells and endothelial cells were investigated on the microfluidic chip.The effect of gradient concentration of VEGF on endothelial cells was studied.Application of this platform could achieve high-throughput cell co-culture research and drug screening;it has the advantages of effectively saving the consumption and labor costs in pre-clinical research.This device has the prospect in studying the interaction between different cells and drug screening.
