Effect Of PRRX1 Expression On VM Formation And Prognosis In Ovarian Cancer

PurposeThis paper mainly studies the paired related homeobox 1(PRRX1)and its pathway molecules expression in ovarian cancer and its relationship with the vasculogenic mimicry(VM)formation,so as to clarify PRRX1 effect on metastasis and prognosis in ovarian cancer.Studying the mechanism of PRRX1 in VM will enhance our understanding of cancer and may provide new therapeutic measures.Methods(1)For the collected 62 cases of ovarian cancer samples conducting PRRX1,Wnt5a,β-cateninimmunohistochemistry staining and CD31/PAS double staining,we observed the expression of these indicators.(2)Analysis the relationship of PRRX1、VM with the clinicopathological and prognosis,and the correlation of PRRX1 and VM、Wnt5a、β-catenin.Results(1)Clinical pathological significance of PRRX1 in ovarian cancerThere was no important difference between PRRX1 and age,tumor size,histological type,degree of differentiation of the tumor(P>0.05).However,there was significant difference in FIGO stage and metastasis(P<0.05).Kaplan-Meier survival analysis showed that the survival time of PRRX1 positive group is significant lower than that of negative group(P=0.038).(2)Clinical pathological significance of VM in ovarian cancerThrough CD31/PAS double staining,we observed VM in ovarian cancer.χ2test showed that there was no distinct difference between VM and age,tumor size,histological type,degree of differentiation of the tumor(P>0.05).However,there was significant difference in FIGO stage and metastasis(P<0.05).Kaplan-Meier survival analysis showed that the survival time of VM exist group is significant lower than that of negative group(P=0.001).(3)The relationship between PRRX1 and VM in ovarian cancerAnalysis on the relationship between the PRRX1 and VM,we found that VM~+in PRRX1 positive group issignificantly higher than VM~+in PRRX1 negative group,and PRRX1 positive expression is related to the existence of the VM(P=0.040).Kaplan-Meier survival analysis showed that the survival time of patients with PRRX1positive expression and VM formation isshorterthan that of PRRX1 negative expression or no VM formation,and has poor prognosis(P=0.002).(4)The relationship between PRRX1 and Wnt5a、β-cateninBased on the immunohistochemical staining of Wnt5a、β-catenin,we observed that PRRX1 overexpression has the relationship withβ-catenin nuclear expression(r_s=0.309,P=0.014)but has no relationship with Wnt5a andβ-catenin cytoplasm expression(r_s=0.110,P=0.396;r_s=0.217,P=0.090).Conclusion(1)As a new EMT inducer,PRRX1 expression is related to the malignant biological behavior,and also affect the survival of ovarian cancer patients.PRRX1 is an independent prognostic indicator in ovarian cancer.(2)VM exists in ovarian cancer,and is related to the malignant biological behavior,and also affect the survival of ovarian cancer patients.PRRX1 affect VM formation.(3)PRRX1 expression has the relationship withβ-catenin nuclear expression,PRRX1regulateVMformationmaybethroughthe classical Wnt/β-catenin signaling,indicating poor prognosis,so PRRX1 might be a candidate marker for the diagnosis and prognosis prediction of ovarian cancer.
