Breast carcinoma is gravely threatened body health of females.Oncogenes highly clustered around breast carcinoma and other cancer cells to promote the development of cancers.Oncogenes that contain G sequence automatically form G-quadruplex,which inversely inhibit the expression of G-quadruplex,utilizing small molecule bind and stabilize G-quadruplex as the clinical cancer treatment are being highly favored.Therefore,to explore the biochemic molecular mechanism of small molecular lays potential valuable clinical significance for cancer treatment.Based on the inhibition activity of Arene Ruthenium complex explored before,the contents of the study included following parts:Firstly,Arene Ruthenium complex RAP07 was synthesized under microwave irradiation and the stuctures of these complexes were characterized by ESI-MS,1H NMR and13C NMR with conformed research results and the theoretical values.Furthermore,corresponding cell biology experiments exploited to study anticancer acitivity and mechanism;MTT assay showed that targeted complex RAP07 selectively inhibited the development of breast carcinoma with an IC500 value of 6.70?M.Further study of flow cytometry and western-blot resulted S-phrase arrest through DNA damaged.Moreover,the result of Western-blot showed RAP07 down-regulate the expression of FANCJ helicase,which lead to DNA damage by blocking DNA replication.Finally,zebrafish experiment showed the in vivo biosafety of RAP07 at a low concentration.Therefore,Arene Ruthenium complex RAP07 inhibited breast carcinoma through double DNA damage anticancer mechanism under a certain biological safety.
