| Objective:This study to test check exercise precondition(exercise precondition,EP)whether can control more complete NLRP3 inflammatory pathway to reduce the content of inflammatory cytokine downstream improve exhaustion exercise induced cardiac function obstacle,and explore what intensity exercise preconditioning has better protective effect on cardiac function.Method:1.Groups and models of experimental animals40 healthy male SD rats are randomly divided into 5 groups(n=8):the control group(CON group),exhaustion group group(EE),low intensity exercise preconditioning and EE group(LEP+EE group),middle intensity exercise preconditioning and EE group(MEP+EE group),High intensity exercise preconditioning and EE group(HEP+EE group).Referring to the sports load standard of Bedford,establish movement in animal models.2.Rat left ventricular myocardial tissue is taken as pathological section.After HE staining,the pathological changes are observed under light microscope.3.Serum reactive oxygen species(ROS),tumour necrosis factor-α(TNF-α),interleuki n-6(IL-6),C-reactionprotein(CRP),Creatine Kinase isoenzyme(CK-MB)are detected by ELISA.4.Use the Millar pressure volume catheter to detect cardiac function,and record the cardiac function cardiac output(CO),stroke volume(SV),end-systolic volume(Ves),end-diastolic volume(Ved),developed pressure(Pdev),end-systolic pressure(Pes),heart rate(HR),ejection fraction(EF),relaxation time constant(Tau),end systolic pressure volume relationship(ESPVR)indicators in each group.5.Western Blot is used to determine the protein expression ofthioredoxin-interacting protein(TXNIP),thioredoxin protein(TRX),nuclear transcription factor kappa B p65(NF-?Bp65),NOD—LRRS containing pyrin domain 3(NLRP3)and cysteinyl aspartate specific proteinase 1(caspase-1)in myocardial tissues of rats.Results:1.Comparison of microstructure of myocardium in ratsCompared with the EE group,the cardiac structure of the LEP+EE,MEP+EE and HEP+EE groups is improved,and the results of the MEP+EE group are more similar to those of the blank group.2.Comparison of serum ROS,TNF-α,IL-6,CRP,CK-MB indexes in ratsCompared with the EE group,LEP+EE,MEP+EE and HEP+EE groups are all decreased,showing statistical significance(P<0.05),and the MEP+EE group is lower than LEP+EE group and HEP+EE group.3.Comparison of cardiac function indexes in ratsCompared with CON group,ESPVR and EF in EE group dropped to the lowest level in the exhaustion group,with a statistically significant difference(P<0.05).The values of CO,SV,Ves,Ved and Tau increased with statistically significant difference(P<0.05).The MEP+EE group is lower than LEP+EE and HEP+EE groups.4.Comparison of myocardial protein TXNIP,TRX,NF-?Bp65,NLRP3,Caspase-1expression in ratsCompared with EE group,LEP+EE,MEP+EE,HEP+EE TXNIP,NF-?Bp65,NLRP3,Caspase-1 the relative expression quantity reduced,the TRX to rise.MEP+EE group TXNIP,NF-?Bp65,NLRP3,Caspase-1 relative expression below the LEP++EE,HEP+EE group,TRX is higher than the LEP+EE,HEP+EE group,with statistical significance(P<0.05).5.Comparison of ROS,myocardial protein,inflammatory factors and cardiac function parameters in ratsROS is positively correlated with NLRP3,caspase-1,CRP and ck-mb(P<0.01).TXNIP is positively correlated with CO,Ved and Tau(P<0.01).NF-?Bp65 and HR,IL-6 into a strong positive correlation(P<0.01).NLRP3 is positively correlated with HR and IL-6,and negatively correlated with EF(P<0.01).Conclusion:1.Exhaustion exercise through increase ROS levels,up regulation TXNIP/TRX/NF-?Bp65/NLRP3 inflammatory signaling pathways,increased NLRP3 inflammatory corpuscle,stimulating downstream inflammatory factor increased,damage the heart function.2.Different intensity exercise preconditioning can cut ROS by reducing oxidative stress,and down-regulation the TXNIP/TRX/NF-?Bp65/NLRP3 inflammatory signaling pathways,reduce NLRP3 inflammatory corpuscle expression,reduce the content of inflammatory cytokines,downstream to improve exhaustion exercise induced cardiac injury.3.Moderate intensity exercise preconditioning is better than low and high intensity exercise preconditioning.It is more in line with the theory of sports medicine and cost benefit to adopt moderate intensity exercise pre-adaptation program as the guidance of military training and athlete training. |
