The Expression Of Rab22a In Breast Cancer And The Correlation With Prognosis Of Survival

Objective:1.To investigate the expression level of Rab22 a in breast cancer tissue,and to analyze the correlation between the expression of Rab22 a and the clinicopathological parameters and prognosis of breast cancer patients.2.To study the expression of Rab22 a in breast cancer cells and the effect of Rab22 a expression on the proliferation of breast cancer cells.Methods:1.From October 2012 to November 2016,210 cases of breast cancer tissues and normal breast tissues were collected from breast surgery department of Sino Japanese Union Hospital of Jilin University.Then we evaluated Rab22 a expression in tumors and normal breast tissues by IHC.At last we compared the differences between Rab22 a expression and age,clinical stage,tumor size,lymph node metastasis,histological differentiation,vascular invasion and OS by statistical methods.2.The expression of Rab22 a in breast cancer cell lines was detected by RT-qPCR,and then the breast cancer cell Rab22 a was silenced and the cell proliferation changes were observed.Results:IHC showed that Rab22 a expression levels were significantly upregulated in breast cancer(P < 0.001),and IHC analysis of 210 paraffin-embedded archived breast cancer tissues showed that high Rab22 a expression was significantly associated with clinical stage,tumor size and lymphatic invasion(P < 0.05).There was no significant correlation between age,pathological differentiation and vascular infiltration(P > 0.05).Univariate and multivariate analysis indicated that Rab22 a was an independent prognostic factor for poorer OS in the entire cohort.In the cytological experiment,the expression of Rab22 a in the breast cancer cell line was significantly higher than that of the normal breast cancer cell line,and the downregulation of Rab22 a expression slowed the proliferation of the cancer cells.Conclusion:Rab22a promotes the proliferation of breast cancer and is related to the metastasis and prognosis of breast cancer,which can be used as an independent prognostic factor for breast cancer patients.
