| The granule of Gehuang Fu Tea is derived from the decoction of traditional Chinese medicine decoction gehuang.It comes from the third volume of zheng zhi hui bu,a famous physician in Qing dynasty.Medicinal tea dosage form of the song "taiping sheng hui fang",the collection of herbal tea.In the previous study,Fu tea was added to radix puerariae decoction in shaanxi province to promote blood lipid and blood glucose regulation in diabetic mice.Based on the literature investigation at home and abroad and the preliminary pharmacological experiments,this study changed the traditional decoction into granule preparation for patients to take and store.To investigate the extraction and molding process of granules from Gehuang Fu Tea,various quality standards,and the protective effects of granules on blood glucose,lipids,liver and kidney in diabetic mice.Study on the preparation of Gehuang Fu Tea granules.The mouse diabetes model was established by animal experiment,and the water extract and alcohol extract of radix astragali of pueraria were given.The results showed that the effects of the alcohol extract group on body weight and blood sugar were significantly better than that of the water extract group.With puerarin and the astragalus glycosides content as quality index,the single factor investigation extraction time,ethanol concentration,extraction times,solid-liquid ratio,again with puerarin,astragalus glycosides content,extract rate,three weighted composite score,the design of four factors three levels orthogonal test for Ge when the best extraction condition was 60% ethanol,8 times,extracting 3 times,each time 1.5 h.Taking the content of tea polyphenols as the quality index of Fu Tea extraction,the effect of extraction time,ethanol concentration,extraction times and material liquid ratio on the extraction rate of Fu Tea was investigated by single factor.Combining traditional tea infusions,retain its taste so choose the response surface method ofwater extraction,the results the best condition for extracting temperature is 88℃,and solid-liquid ratio of 1:48,extracting time,92 min,tea polyphenols extraction yield of 12.43%.To granule molding moisture absorption rate,key quality indicators of forming rate,melt examination,selection diluent kinds,proportion and dry paste and wetting agent concentration,dosage proportion,to Gehuang tea extract powder particles,microcrystalline cellulose,eight dextrin 3:2:1 dosage proportion,90%ethanol as wetting agent,dosage of 15% of the extract powder dosage,55℃ dry,whole grain of 14 mesh,quick.Quality control study.According to the quality requirements of particle dosage form in the Chinese pharmacopoeia published in 2015,the test of granule size,solubility and moisture of the finished products of Gehuang Fu Tea meet the pharmacopoeia specifications.TLC was used to identify radix puerariae and radix astragali.To establish a HPLC method for the determination of puerarin and astragaloside in finished products.The critical moisture content obtained by saturated solution method is 69% for reference in storage environment.Study on hypoglycemic effect 30 mice were fed high-fat and high-sugar diets for 4weeks and were given 150mg/kg STZ intraperitoneally after fasting for 12 hours to build a diabetic mouse model.Start experiment points,high fat and sugar the blank group,Gehuang Tea eight protection group and metformin group,the injury group,lavage,during measuring blood glucose,body weight,diet quantity of drinking water,8 weeks after death,take heart,liver,kidneys and viscera index weight calculation,take the index,measuring the serum lipid in serum and liver kidney index(Alt,aspertate aminotransferase,urea nitrogen,creatinine);The indexes of oxidative stress(superoxide dismutase,catalase,malondialdehyde)in the centrifugal fluid of the two tissues were measured by homogenating liver and kidney tissues.Results:after 8 weeks of drug administration,compared with the injury group,the blood glucose was significantly reduced,body weight was increased,and the amount of drinking water and diet was decreased.TG,TC and LDL levels of the protective group were significantly lower than those of the injury group(P<0.01,P<0.05,P<0.05),and HDL levels weresignificantly higher than those of the injury group(P<0.01).The indexes of liver and kidney(Alt,ast,urea nitrogen and creatinine)were significantly reduced(P<0.01).Higher SOD and CAT values indicated increased antioxidant capacity,lower MDA values indicated decreased oxidation reaction,and higher SOD and CAT values in renal tissue were compared with the injury group(P<0.01,p<0.05),and lower MDA values(P<0.01,P<0.01).The liver tissue SOD and CAT values were increased(P<0.01,p<0.05)and MDA values were decreased(P<0.01,P<0.01).The results indicated that the granules of gehuang Fu Tea had certain regulating effects on blood glucose reduction and lipid metabolism in mice,and had protective effects on liver and kidney. |
