Background and objective:This study was designed to detect the expression of dystroglycan(DG)in primary and metastatic lesions of breast carcinoma and to analyze the relationship between DG expression and clinicopathologic characteristics.The potential mechanisms of DG in the development of breast cancer were discussed.Methods:A total of 255 cases of breast cancer were collected,including 130 patients with axillary lymph node metastasis and 125 without axillary lymph node metastasis.Immunohistochemical staining was used to detect the expression of DG subunit(alpha-DG,beta-DG)in paraffin samples of adjacent breast tissue,primary breast cancer and lymph node metastasis.The differences in expression and the relationship between DG expression and clinicopathologic characteristics were analyzed.Results:Between January 2010 and December 2010,130 cases of female breast cancer patients with complete medical records were confirmed by pathology of the existence of axillary metastatic lymph nodes,125 cases were confirmed without axillary lymph node metastasis breast cancer.All patients underwent radical mastectomy.The results showed that the positive expression rate of alpha-DG in adjacent breast tissue,breast cancer,and axillary lymph node metastasis was decreasing.There was no significant difference in the positive expression rate of P-DG in the adjacent breast tissue,primary breast cancer,and lymph node metastasis.There was no significant difference in positive expression rate between different T stages,N stages and molecular typing.The positive expression rate of alpha-DG in non-metastatic breast cancer was higher than that of metastatic breast cancer.Conclusion:1.The positive expression rate of a-DG in normal breast tissue,primary breast cancer and lymph node metastasis is decreased,and the difference was significant,which suggesting that it may be related to the occurrence,development and metastasis of breast cancer.2.There is no significant difference in the positive expression rate of a-DG in different T stage,N stage and molecular type tumor tissues.3.Loss of a-DG expression in primary breast cancer may suggests a higher risk of axillary lymph node metastasis.
