Objective: To explore the expression level of plasma miRNA-214 expression difference in patients with cervical cancer.And to analyze its effects on the proliferation’ cell cycle and apoptosis of cervical cancer cells.Materials and Methods: From January 2016 to January 2018,50 patients with invasive cervical cancer diagnosed by cervical biopsy pathology in Hefei Tumor Hospital of Chinese Academy of Sciences were collected as experimental group.The average age was(56.2 +14.3)years,and there were no other gynecological diseases or chronic diseases.Exclusion criteria: Non-first-diagnosed patients,i.e.those who have been diagnosed as cervical cancer but have received surgical treatment,radiotherapy and chemotherapy,have a history of severe organ diseases and other systemic tumors,such as heart,liver,lung,brain,kidney and other organ and blood system diseases.In addition,50 healthy women with negative blood pressure,laboratory tests,gynecological examinations,ultrasound and cervical cancer screening were selected from Hefei Cancer Hospital of Chinese Academy of Sciences.The average age was(50+7.9)years.Those who excluded other chronic diseases or malignant tumors were taken as normal control group.Three days before operation and on the day of consultation,healthy volunteers collected 2 ml blood samples from veins containing EDTA anticoagulant.After shaking gently,the samples were stored at 4 C,1500 rpm and centrifuged for 15 min.The supernatant was kept at-80 C for reserve.Real-time fluorescence quantitative PCR was used to detect and analyze the expression of microRNA214 in plasma of the two groups,and the difference was compared.MiRNA-214 mimics were transfected by liposome-mediated transfection.The proliferation,cell cycle and apoptosis of cervical cancer cells were detected by CCK-8and flow cytometry.Result: 1.The relative expression of microRNA214 in cervical cancer and controlgroup was different after real-time fluorescence quantitative PCR detection.The level of microRNA214 in cervical cancer patients’ plasma(1.01(+0.041 pg/ml)was significantly lower than that in normal control group(2.96(+0.67 pg/ml).The difference between the two groups was significant.2.To study and analyze the microRNAs in cervical cancer patients’ plasma.The relationship between the level of-214 and the pathological data of cervical cancer patients(including lymph node metastasis,tumor size,vascular invasion,FIGO stage and the degree of tumor differentiation)revealed that the expression of microRNA-214 in plasma of patients with cervical cancer significantly decreased when the pathological findings of patients with cervical cancer after operation indicated the presence of lymph node metastasis and vascular invasion.The difference was statistically significant(P=0.0231,0.0295;P<0.05).There was no significant difference in the expression level of microRNA-214 in plasma of patients with other pathological data(including tumor size,FIGO stage and tumor differentiation degree)(P > 0.05).3.CCK-8 method was used to detect the effect of microRNA-214 expression on the proliferation of SiHa cells.It was found that the growth rate of SiHa cells was inhibited in the microRNA-214 mimetic transfection group.4.Compared with the control group,the proportion of cervical cancer cells in S phase and apoptotic cells in G2/M phase in the microRNA-214 group decreased significantly(P < 0.05),while the proportion of apoptotic cells in G2/M phase decreased significantly(P < 0.05).The results showed that the decrease of the expression level of microRNA-214 in cervical cancer cells might inhibit G2/M arrest,whereas the over-expression of microRNA-214 could significantly promote the apoptosis of cervical cancer cells.Conclusion: 1.The expression of microRNA-214 is down-regulated in cervical cancer tissues,which has certain accuracy as a molecular marker for early diagnosis of cervical cancer.2.MicroRNA-214 can inhibit the entry of cervical cancer SiHa cells into S phase and promote the apoptosis of SiHa cells.MicroRNA-214 may play a role by inhibiting the proliferation,migration and invasion of cervical cancer cells.It has theeffect of inhibiting cancer.
