Objective:Using gelatin aqueous solution as raw materials.The preparation of a peptide-iron chelating compound with blood reactivity was processed by protease and ferrous chloride?FeCl2·4H2O?.In addition,the preliminary pharmacodynamic study and stability of peptide-iron complex were studied.Methods:The enzymatic hydrolysis process of gelatin bromelain and trypsin was studied by degree of hydrolysis and molecular weigh;The hematopoietic model of mice and leukocyte reduction model of cyclophosphamide induced by bloodletting were used to investigate the effect of gelatin and its enzymatic hydrolysate on enrich the blood and hemameba.Preparation of peptide-iron complex was studied by the index of the Chelation rate and molecular weigh.The iron-deficiency anemia mice model was used to investigate the blood enriching effect of peptide-iron complex.The stability of gelatin peptide-iron chelate was studied by influencing factors and heating accelerating method.Results:Process of the gelatin peptide-iron chelate is:Bromelain digestion pH=7,the amount of enzyme bromelain5%,temperature 55?,pepsin digestion time is 1.5 h,trypsin digestion pH=7.5,the amount of enzyme bromelain 2%,temperature 40?,pepsin digestion time is 3 h,boiling for 15min,cryopreserve?04??for 6h,then take supernatant upper sample after Centrifugation?4500 r/min,10 min?,the amount of eascorbic acid 0.1%,pH=5,the amount of Ferrous ion is10 mg?mg/1 g gelatin?,and mixing 20 min with a magnetic whisk at temperature 20?,then centrifuged to remove precip;Gelatin peptide can make the loss of blood caused by blood deficiency mice model of Hb and RBC were significantly improved,and white blood cells in mice caused by cyclophosphamide reduced model of the WBC level improved significantly;The gelatin peptide-iron chelate can significantly increase the values of HGB,HCT,MCV,MCH,MCHC and CHCM in blood of mice with iron deficiency anemia,and decrease RDW and HDW values;The molecular weight distribution of the donkey-hide elatin and fractions ranged from 168 Da to less than 1.7×105 Da.Gelatin hydrolysate and eptide-ironchelatefractions ranged from 150Da to less than 5×104Da,and ranged mostly from 2000 Da with narrow distribution;3 months stability test shows each index comply with the requirements.Conclusion:The prepared gelatin peptide-iron compound has stable technology and has significant improvement hypoferric anemia.
