objective: Diabetic retinopathy(DR)originates from non-proliferative or proliferative retinal vascular abnormalities.The main characteristic of DR is that hyperglycemia damages retinal microvascular system and leads to compensatory neovascularization.In recent years,the protein-coding genes related to hyperglycemia have been paid attention to in the progress of DR,especially micro RNA(micro RNA,micro RNA,micro RNA)has become the current research hotspot.Micro RNAs have been shown to play a key role in the signaling pathways of endothelial cells and perivascular cells.It is mainly involved in the proliferation,migration and apoptosis of retinal cells and the formation of DR-related neovascularization,and plays a role in regulating angiogenesis.These findings suggest that there may be one or more specific serum micro RNAs expression patterns in DR,which can be used as a new non-invasive method for early prediction of the occurrence and development of DR.In this study,we detected the different expressions of six kinds of micro RNAs in diabetes mellitus(DM)and diabetic retinopathy(DR)patients,including micro RNA21,micro RNA200 b,micro RNA126,micro RNA221,micro RNA181 c and micro RNA195,to explore the expression and significance of serum micro RNAs in diabetic retinopathy(DR).Methods : Sixty consecutive cases of type 2 diabetes mellitus(DM)hospitalized in the Department of endocrinology of Mianyang Central Hospital from January to December 2017 were selected as control group(DM group),and 60 consecutive DR patients in ophthalmology clinic were selected as experimental group(DR group).The diabetic retinopathy in the DR group was in accordance with the clinical diagnostic criteria of DR established by the 1984 Academic Conference on fundus diseases.Two groups of patients were examined by the same senior ophthalmologist.After mydriasis,fundus examination was performed under preoptic lens,fundus photography was improved,and fundus fluorescein angiography was performed.No diabetic retinopathy was found in DM group,and diabetic retinopathy was found in DR group.Quantitative Real Time-Polymerase Chain Reaction(q PCR)was used to detect the expression levels of serum micro RNAs-21,micro RNAs-200,micro RNAs-126,micro RNAs-221,micro RNAs-181 c and micro RNAs-195.QPCR,also known as real-time fluorescence quantitative PCR.Statistical software SPSS20.0 was used to analyze the difference of the expression levels of micro RNAs between the two groups.ROC curve was used to evaluate the sensitivity of micro RNAs in the diagnosis of DR.And specificity.Results: Quantitative Real Time-Polymerase Chain Reaction was used to detect the expression of six kinds of micro RNAs in serum of DM and DR groups,and the relative expression was calculated by 2-??Ct.The formula is: relative quantitative formula?? Ct = ?Ct(micro RNA)-?Ct(U6).And through the transformation of log2(x),-??Ct is obtained.After data processing and analysis,we found that the expression of micro RNA200 b,micro RNA21 and micro RNA221 in serum of DR patients was higher than that of control group,while the expression of micro RNA126 was lower than that of control group(P< 0.01).The expression of micro RNA181 c in serum of DR patients was up-regulated,but the difference was not statistically significant(P > 0.05).The expression of micro RNA195 in serum of DR patients was down-regulated,and the difference was not statistically significant(P > 0.05).The areas under the ROC curves of micro RNA-21,micro RNA-200 b,micro RNA-126 and micro RNA-221 were 0.878,0.974,0.916 and 0.817.Conclusion:The expression of micro RNA-21,micro RNA-200 b and micro RNA-221 in serum of DR patients was significantly up-regulated,and the expression of micro RNA-126 was significantly down-regulated.Detecting the expression of serum micro RNA-21,micro RNA-200 b,micro RNA-126 and micro RNA-221 may be related to the occurrence and development of DR,providing a new research direction for early diagnosis and targeted therapy of DR.At the same time,the area under the ROC curve of micro RNA-21,micro RNA-200 b,micro RNA-126 and micro RNA-221 suggests that serum micro RNA-200 b has the highest sensitivity and specificity for the diagnosis of DR,which provides a new way for early diagnosis and evaluation of DR.
