| Objective:By comparing the content of active ingredients in Chelidonium majus L from different areas and harvesting time in Jilin Province,the effects of harvesting time and area on the content of active ingredients in Chelidonium majus L were studied.By replicating animal models of cough-inducing mice and asthma-inducing cavy,analysis of the effect of the content change of active ingredients in Chelidonium majus L on it’s pharmacodynamics,clarify the correlation between the areas,harvesting time and the content of active ingredients and pharmacodynamics of Chelidonium majus L in Jilin Province,and to guide the rational use of Chelidonium majus L in clinic.Methods:Determination of Chelerythrine in Chelidonium majus L from Different areas and Harvesting time in Jilin Province by High Performance Liquid Chromatography,establishment of cough model induced by ammonia in mice and asthma model sensitized by cavy ovalbumin,the cough latency,cough frequency,levels of inflammatory factors interleukin-1 and interleukin-6 were observed in mice of each group,the latency of asthma,behavioral score,airway mucopolysaccharide content,CD4+/CD8+,and lung histopath-ological changes were observed in cavy each group.Results:There were differences in the content of chelidonine between the samples of Chelidonium majus L from different areas and harvesting time,among 54 samples of Chelidonium majus L,28 samples contained less than 0.02%chelidonine,which did not meet the standard of Chinese Pharmacopoeia in 2015edition.Changchun,Baicheng,Yitong,Jilin,Jiutai,Shuangyang,Tonghua,Fuyu and Jingyu groups all prolonged the latency of cough in mice and decreased the number of coughs within 3 minutes(P<0.05 or P<0.01),the latency of asthma in cavy was prolonged and the behavioral score was decreased(P<0.05 or P<0.01).The concentrated liquor of Chinese Chestnut produced by Fuyu and Jingyu could reduce the release of inflammatory mediators and inhibit the chronic airway inflammation in mice(P<0.05).The concentrated solution of Chelidonium majus L from Fuyu and Jingyu can reduce the release of inflammatory mediators in mice,the effect of inhibiting chronic airway inflammation was significant(P<0.05).Changchun,Yitong,Jilin,Jiutai,Shuangyang,Tonghua,Fuyu and Jingyu could significantly reduce the content of mucopolysaccharide in the airway of cavy,and increase the value of CD4+/CD8+(P<0.05 or P<0.01),among them,taking Chelidonium majus L from Jingyu was better than taking Changchun,Baicheng,Yitong,Jilin,Jiutai,Shuangyang,Tonghua and Fuyu(P<0.05 or P<0.01)in controlling the latency of cough in mice,cough times in 3 minutes,latency of asthma in cavy,behavioral status of asthma,content of airway mucopolysaccharide and CD4+/CD8+.HE staining results showed that the infiltration of perivascular inflammatory cells in lung tissue of cavy in Changchun,Jilin,Shuangyang,Tonghua,Fuyu and Jingyu group was significantly less than that in model group.Conclusion:Harvesting time and areas all affect the content of active ingredients in Chelidonium majus L in Jilin Province,the most effective ingredients were collected in Jingyu area in July.Changes in the content of active ingredients in Chelidonium majus L will affect the effect of relieving cough and asthma.Chelidonium majus L has certain advantages in treating cough and asthma symptoms in Jingyu area of Jilin Province. |
