ObjectiveTo investigate the inhibitory effect of resveratrol?RES?on apoptosis of human retinal pigment epithelial cells induced by sodium iodate?NaIO3+RES?in vitro and in viv.MethodsIn vitro experiments,RPE cells were injured by sodium iodate to simulate dry macular degeneration in human retinal pigment epithelium-19?ARPE-19?cell lines.RPE cells were divided into three groups:blank group,sodium iodate injury group and experimental group containing resveratrol.The morphology of RPE cells in each group was observed and recorded.MTT assay was used to detect the optimal concentration of RES on RPE cells and the fine protective effect of RES on RPE cells damaged by different concentrations of sodium iodate.The expression of apoptotic proteins Bax and Bcl-2 was detected by Western blot.Hoechst 33342 and flow cytometry were used to detect the difference of total apoptotic cells and the types of apoptotic cells in each group.In vivo experiments,30 male C57BL/6J mice were randomly divided into control group,model group?NaIO3?and experimental group?NaIO3+RES?.On the 1st,7th,14th,21st and 28th days after NaIO3 was injected into tail vein of rats,fundus photography and OCT imaging were performed,and eyeball tissues were taken for HE staining to observe the changes of retinal structure.ResultsIn vivo experiment:On the 7th day after NaIO3 injection,OCT retinal thickness in the experimental group was thicker than that in the model group,and RPE layer,photoreceptor and outer nuclear layer were less damaged than that in the model group.In vitro experiments:RES has a strong protective effect on RPE cells at the concentration of 5?m·L-11 and has a strong protective effect on RPE cells with different degrees of injury.Relative expression of Bax protein?0.45±0.07?in sodium iodate group with RES was significantly lower than that in sodium iodate group?0.90±0.09?and the difference was statistically significant?P<0.05?.There was no significant difference between sodium iodate group?0.34±0.06?and blank controlgroup?P>0.05?.Relative expression of Bcl-2protein in sodium iodate injury group with RES?0.56±0.14?was significantly higher than that in sodium iodate injury group?0.24±0.04?.The difference was significant,?P<0.05?.but there was no significant difference compared with the blank control group?0.73±0.08?.The results of Hoechst 33342 showed that the content of apoptotic cells in the sodium iodate group with RES was significantly lower than that in the sodium iodate group.Flow cytometry showed that the number of apoptotic cells in the sodium iodate group with RES was less than that in the sodium iodate group with RES.ConclusionsThis experiment proves that RES has a good inhibitory effect on the apoptosis of RPE cells injured by sodium iodate,and has a better protective effect on retinal damage in the model of AMD simulated by sodium iodate,which has potential value for the treatment of AMD.
