Role Of JMJD2D In Citrobacter Rodentium Infection

Histone demethylase JIMJD2D can remove methyl residues from H3K9me2/3 and active the expression of genes.Previously,It reported that JMJD2D was associated with the tumorigenesis,could recuit to DNA damage site,facilitate the DNA repair and was essential for the initiation of DNA.C.rodentium is highly pathogenic to weanling mice and its mechanism is similar to EPEC/EHEC.But JMJD2D's role in the mice infected with C.rodentium is unknown.This study discusses the function of JMJD2D in the C.rodentium infection mode.Firstly,We treated WT mice and JMJD2D-/-mice with C.rodentium by gavage.Interestingly,JMJD2D was up-regulated on enterocyte sections from the colon tissue?suggesting that JMJD2D might play a role in C,rodentium infection.Ulteriorly,JMJD2D-/-mice impaired clearance of C.rodentium,losed more weight and suffered from more damage in the colon compared with the WT mice,showing that up-regulated JMJD2D could be beneficial to the host infected with Citrobacter rodentium.Meanwhile,the mRNA level of IL-6,TNF-?,IL-1? were up-regulated in different degrees,but the mRNA level of ?-defensionl-4 of JMJD2D-/-mice were significantly decreased at last point compared with the WT mice.In conclusion,JMJD2D could reduce the level of inflammatory cytokines and promote the secretion of antimicrobial peptides.Further,the mRNA level of IL-17F in JMJD2D-/-mice was downregulated at day 14 compared with wild-type mice,but the IL-17A level was unchanged.The result showed that JMJD2D could regulated the expression of IL-17F,but not IL-17A.All together,JMJD2D might regulate antimicrobial peptides through IL-17F,finally,protect the host from C.rodentium and it can also provide new ideas for the treatment of bacterial enteritis.
