Liquid biopsy is a low-invasive tumor detection technology.The sample source can be various body fluids of the human body.This technology has the advantages of convenience,low risk and strong applicability.Compared with traditional tumor detection technology,it can treat cancer.Early screening,real-time monitoring,efficacy assessment,and prognosis prediction.Most of the detection targets are tumor cells and related products(protein,DNA,RNA).This experiment mainly uses microfluidic chips to optimize and improve the tumor separation steps in the existing liquid biopsy.Microfluidic chip is a microfluidic mechanics and design technology that integrates many chemical and biotechnologies into a microchip of several square centimeters to implement complex operational steps in a micron-scale chip-channel network.Known as the "chip lab",it is widely used in biochemistry and medical scenarios,and is characterized by miniaturization,integration and convenience.At present,the use of microfluidic technology to separate cells can be divided into "label capture" and "label-free capture".The former has low cost and high efficiency,but has poor specificity and low purity;the latter has high specificity but high cost.In this experiment,the two experimental ideas are combined,and the concept of"tag-gating" is proposed.The microfluidic chip technology is used to filter the cells that pass into the sample using the "liquid filter" structure,and the labeled tumor cells are isolated and blocked.Control the tag cells.In this experiment,the structural design of the liquid filter chip is also completed.The material is made of acrylic plate.The chip itself can be reused many times,and the immune capture is added at the same time,which avoids the problem of low specificity of the label-free capture method and reduces the cost.
