ObjectiveTo analyze the expression of Axin2 in SNU-245 and SNU-308 cell lines and bile duct carcinoma tissues was analyzed to explore its role in the pathogenesis of bile duct carcinoma and its mechanism.MethodsIn this study,stabled bile duct cancer cell lines SNU-245 and SNU-308 were established by pLKO-Tet-ShAxin2 plasmid transfection.Proiiferation ability of cancer cells was detected by cell proliferation assay,migration ability of cancer cells was detected by cell scratch experiment,invasion ability of cancer cells was detected by transwell assay,and Axin2 and Snail protein expressions were detected by western blot Also collected 116 samples of cholangiocarcinoma,which was followed up for 5 years.The expression of Axin2 and Snail were observed by immunohistochemical staining,and their correlation with the clinicopathological features of bile duct carcinoma was analyzed.Results(1)In SNU-245 and SNU-308 cell lines of human bile duct carcinoma,Axin2 knockdown significantly reduced cell proliferation,migration,invasion and Snail expression.Axin2 protein was highly expressed in 59 cases(50.9%)and Snail protein in 54 cases(46.6%)of 116 patients with cholangiocarcinoma,and the expressions of the two proteins were positively correlated(P<0.001).(2)The survival rate of patients with high Axin2 and Snail protein expression was lower(all P<0.001).Multivariate analysis showed Axin2 protein(95%confidence interval,1.569-5.380;P=0.001)and Snail protein(95%confidence interval,1.609-6.186;P=0.001)high expression was an independent risk factor for poor prognosis of patients.Combined with the clinical and pathological characteristics of patients,the prognostic model of Axin2 and Snail expression was established.Conclusion(1)Axin2 plays an important role in the proliferation,migration and invasion of bile duct cancer cells,and its mechanism is related to the high expression of Axin2 to maintain the stability of Snail.(2)Axin2 can be used as a marker for the diagnosis,target treatment and prognosis of cholangiocarcinoma,and is closely related to poor prognosis of patients.
