PurposeAdiponectin is an adipocyte-derived hormone,which can improve insulin resistance and reduce the risk of type 2 diabetes.Studies in recent years found that adiponectin also plays an important role in the regulation of anxiety and depressive-like behavior.However,the underlying mechanism of adiponectin on depressive and anxiety like behavior remains elusive.In this study,we linked serotonin system to the regulation of adiponectin on anxiety-like and depression-like behaviors.On the other hand,serotonin(5-HT)is an important monoamine neurotransmitter in the central nervous system,which most definitely originated in the raphe nucleus,and mainly involved in emotion regulation,cognitive function(learning and memory),appetite and sleep.A large number of studies have proved that 5-HT is closely related to depression-and anxiety-like behaviors,particularly serotonin hypofunction hypothesis on the regulation of depression is generally recognized.The aim of this study was to investigate whether adiponectin receptor AdipoRl on 5-HT neurons has effects on depression-and anxiety-like behaviors in mice.It is worth mentioning that sexual gender plays an important role on behavior,so we performed experiments on female and male mice respectively.MethodsThe AdipoRlflox/flox mice and ePetl-cre mice were intercrossed to generate AdipoRlflox/flox mice/ePetl-cre mice,in which AdipoRl in the 5-HT neurons have been Knockout.depression-like behaviors and anxiety-like behaviors were assessed after 8 weeks old,such as the female urine sniffing test smell,the Elevated plus maze,sucrose preference test,Forced Swimming test,Tail Suspension test and Learned Helplessness.The tests were performed by sexual gender,the final data can tell the behavior changes of female mice and male mice.Result1.In the assessment of depression-like behavior experiment,we found AdipoR1flox/flox mice/ePetl-cre male mice takes less time of smelling the female urine in female urine sniffing test and also less time of immobility in forced swimming test.While in other depression behavior test,such as sucrose preference test,tail suspension test,it indicated no significant difference between the two group.As to the female mice,we found AdipoR1flox/flox mice/ePetl-cre female mice takes more time of immobility in forced swimming test.While in other depression behavior test,such as sucrose preference test,tail suspension test,it indicated no significant difference between the two group.2.In anxiety-like behavior test,there is no difference in the percentage of open arm entries and spending time in the open arms between the two group.As to the female mice,there is still no difference in the percentage of open arm entries and spending time in the open arms between the two group.ConclusionsBy the above results,it suggested that the male and female mice of knockout AdipoRl on 5-HT neurons can induce the depression-like behavior,but there is no effect on anxiety-like behavior.
