Abstract/Summary:
Objective This article investigates the relationship between the level of PCSK9 in serum and the blood fat(or other atherosclerosis risk factors)for T2 DM patients.We also explore the correlation between PCSK9 and the secretion of inflammatory factors(such as IL-6,IL18)in the pathogenesis of atherosclerosis.Methods Using the bridging ABC-ELISA method to detect 60 cases of T2 DM patients and 28 cases of normal control group in serum general indicators,including lipid,fasting blood-glucose,cholesterol(TC),triglyceride(TG),low density lipoprotein(LDL),high-density lipoprotein(HDL),glycosylation hemoglobin(HbA1c).For 60 patients with T2 DM double carotid artery and cerebrovascular color doppler ultrasound,including the right common carotid artery intima-media thickness(RCCA IMT),the right internal carotid artery intima-media thickness(RICA IMT),the left common carotid artery intima-media thickness(LCCA IMT),the left internal carotid artery intima-media thickness(LICA IMT),average total artery intima-media thickness(MCA IMT),inner diameter of RCCA,inner diameter of RICA,inner diameter of LCCA,inner diameter of LICA,the right common carotid artery resistance index(RCCA RI),the right internal carotid artery resistance index(RICA RI),the left carotid artery resistance index(LCCA RI),the left internal carotid artery resistance index(LICA RI),the maximum blood flow velocity of the right carotid artery(RCCA Vmax),the maximum blood flow velocity of the right internal carotid artery(RICA Vmax),the maximum blood flow velocity of the left carotid artery(LCCA Vmax),the maximal blood flow rate of the left carotid artery(LICA Vmax),the miximal blood flow rate of the right carotid artery diastolic velocity(RCCA Vmix),the maximal blood flow rate of the right internal carotid artery diastolic velocity(LCCA Vmix),the maximal blood flow rate of the left cervical artery diastolic velocity(LCCA V mix),and the maximal blood flow rate of the left internal carotid artery diastolic velocity(LICA V mix),etc.It was found that that correlation between PCSK9 level and the risk factor of atherosclerosis in type 2 diabetes mellitus were investigated by use the methods of bridge abc-elisa to detect the levels of PCSK9 and the levels of IL-6 and IL18,compare the correlation between the two groups,the correlation factors of serum PCSK9 level in T2 DM group were analyzed by person correlation and multifactor linear regression.Results 1.General situation: compared with the healthy group,the blood lipid,blood glucose,cholesterol(TC),triglyceride(TG),low density lipoprotein(LDL),high-density lipoprotein(HDL),glycosylated hemoglobin(HbA1c),PCSK9,IL-6,and IL-18 concentration were increased,and the difference was statistically significant(P<0.05);2.Compared with patients with type 2 diabetes without vascular disease(group A),serum lipids,blood glucose,cholesterol(TC),triglyceride(TG),low density lipoprotein(LDL),high-density lipoprotein(HDL),glycosylated hemoglobin(HbA1c),PCSK9,IL-6 and IL-18 increased in serum of patients with type 2 diabetes mellitus(group B),and the difference was statistically significant(P<0.05).3.The serum PCSK9 levels in the control group,group A and group B were significantly positively correlated with the common risk factors of atherosclerosis,such as SBP,TC,LDL-C,TG,FBG and other atherosclerosis,and had A statistically significant negative correlation with HDL(p < 0.01),and had no correlation with fasting c-peptide,FINS,HOMA-IR,HOMA-B(P>0.05).4.The serum PCSK9 level of type 2 diabetes mellitus was significantly positively correlated with the right common carotid artery intima-media thickness(RCCA IMT),the right internal carotid artery(LCCA IMT),the left carotid artery endothelial thickness(LCCA IMT),the left internal carotid artery(LICA IMT),and the average total arterial thickness(MCA IMT)(P < 0.01).It was found that that correlation index of carotid atherosclerotic such as internal diameter of RCCA,internal diameter of RICA,internal diameter of LCCA,inner diameter of LICA,RICA V Max,LICA R I,RICA R I,LCCA RI,and the like were also positively correlate(P < 0.05),and showed a marked negative correlation with RCCA Vmax,LCCA Vmax,LICA Vmax,RCCA V mix,RICA Vmix,LCCA Vmix,LICA Vmix,etc.(P< 0.05).5.There was a significant positive correlation between PCSK9 level in serum of type 2 diabetic patients and IL-6 and IL18(P<0.01).Conclusions The increase of PCSK9 in serum of patients with type 2 diabetes is one of the independent risk factors of atherosclerosis,which accelerates the development of diabetic atherosclerosis together with IL-6 and IL-18.
