Objective: Intravascular therapy is one of the main treatments for the arterial occlusive disease with its unique advantages of minimally invasive.However,intravascular restenosis after endovascular intervention is an unavoidable problem.Our study group intends to use nuclear transfection to make EPCs over-express Periostin protein,then,the mice model of carotid artery injury was treated with cell transplantation and the effect was evaluated.We are working to find a better treatment for restenosis after endovascular intervention.Methods: ApoE-/-mice were fed on high-fat fodder for four months.Before and after feeding,the mice were weighted and the blood lipid levels were detected.After some mice were killed,HE staining of there carotid artery and live were observated.We purchased EPCs that stably express Periostin protein and cultured.Then,the normal EPCs and over-express Periostin protein EPCs were injected into mice through the tail vein.After four months,the re-endothelialization of injured carotid artery was detected by Evans blue staining.The size of atherosclerotic plaque area in the injured carotid artery were measured by HE and Red Oil O staining.The determination of Ki67 and PCNA expression were measured by immunohistochemical staining.Results: The weight and blood lipid of the mice increased after four moths feeding,pathological staining observation of their carotid artery and liver showed that the tissue structure had changed a lot.The results indicated that the injured carotid artery mice model was established successfully.We cultured EPCs in vitro from bone marrow source.Cell morpHology was observed by microscope and the picture showed that EPCs have a pebble-like shape.The expressions of CD34 were detected by immolunohistochemical technique and they were positive.The expression of the Periostin vector plasmid was verified in EPCs by Western blot analysis and it was positive.The injured carotid artery mice were treated by over-express Periostin protein EPCs over-express respectively.Conclusion: In our study,The injured carotid artery mice model was established successfully.The over-express Periostin protein EPCs of mouse were cultured in vitro.Our results indicated that the effect of over-express Periostin protein EPCs is better than the other's group for cell therapy.
