Objective:For providing theoretical basis of clinical treatment of NAFLD using traditional Chinese medicine,it is to further elucidate the molecular mechanism of intervention of Jiangan Xiaozhi prescription,which is taking blood circulation and removing blood stasis as the treatment principle,in the treatment of NAFLD insulin resistance by observing animal model ROS activity.Methods : Rats were fed with high-sugar and high-fat diet.After 30 days,hepatic lipids were measured and 6 were randomly selected for pathological section.NAFLD model Rats were randomly divided into pathological model high-fat group,metformin group(0.09g/kg),Jiangan Xiaozhi prescription low-dose group(5.43g/kg crude drug),middle dose group(10.86g/kg crude drug),High dose group(21.73 g/kg crude drug),15 in each group.The groups were given the corresponding test drugs by intragastric administration at the same time of feeding;the blank group and the high-fat group were given the same volume.Fasting insulin levels and fasting blood glucose levels were measured after 60 days,and insulin resistance index(HOMA-IR),liver reactive oxygen species(ROS)levels,and liver lipid deposition were calculated.Results:Compared with normal rats,fasting insulin content,HOMA-IR index and ROS level were significantly increased in the model group(P<0.05).Liver steatosis was significantly enhanced(P<0.05).Jiangan Xiaozhi prescription high-and medium-dose group and metformin could inhibit ROS and improve insulin resistance in rats(P<0.05,P<0.01).Conclusion : Jiangan Xiaozhi prescription is effective for NAFLD,and its mechanism may be related to inhibition of ROS and improvement of insulin resistance.
