| Objective:To explore the distribution of single nucleotide polymorphisms and the phenomenon of linkage disequilibrium in 2 SNPs of Wingless-Type MMTV Integration Site Family Member 10A(WNT10A)Gene between Uyghur and Han nationalities in Xinjiang.Methods:Genotypes and allele frequencies of 2 SNPs were examined by PCR technique and sequencing method,combined with DNA STAR and Chromas 2.0 software to carry out.The data were statistically analyzed by the software SPSS 23.0.At the same time,the linkage disequilibrium(LD)results were evaluated by Haploview 4.1 software.Results:1.The distribution about genotypes of rs10177996(X~2=6.165,p<0.05)in Uygur and Han populations in Xinjiang were statistically significant.The homozygous type CC of rs7349332 was dominant in Uygur and Han populations,but there was no ethnic difference(X~2=0.263,p>0.05).2.The distribution of associated alleles in rs10177996(X~2=4.556,p<0.05)were statistically significant in the population of Uygur and Han nationalities in Xinjiang.However,rs7349332(X~2=0.229,p>0.05)did not show ethnic differences.3.There were statistical differences in comparison of genotypes distributions of rs10177996 in Xinjiang Uygur and Han populations(p<0.05),Xinjiang Uygur and European populations(p=0.05),Xinjiang Han and European populations(p<0.01),and the proportion of CC,CT,and TT on Uygur individuals of the sample population(8.21%,30.6%,and 61.19%,respectively)was between the Han population(9.74%,43.51%and46.75%,respectively)and the European(2.64%,30.09%and 67.27%,respectively).In addition,the distribution ofC/T alleles was statistically different between the Han population and the European(X~2=19.483,p<0.01),nevertheless,there was no statistical significance in the distribution of Xinjiang Uygur and European populations.(X~2=3.566,p>0.05).4.Linkage Disequilibrium analysis showed that there was a significant linkage disequilibrium between rs10177996 and rs7349332(D’=0.956,r~2=0.687)of Han population in Xinjiang.The relationships about linkage disequilibrium between rs10177996 and rs7349332(D’=0.597,r~2=0.313)of Uygur population,and of Uygur and Han populations(D’=0.788,r~2=0.501)in Xinjiang can be seen.Conclusion:The distribution of single nucleotide polymorphisms about WNT10A Gene rs10177996 of Xinjiang Uygur population,Xinjiang Han population,and European are different.The linkage disequilibrium existed between rs10177996 and rs7349332.This study provides genetic data about 2 SNPs of Uygur and Han populations in Xinjiang,and lays a theoretical foundation for the development of dentition recognition and identification between the two ethnic groups. |
