Objective:To explore the effects of microRNA-24 on proliferation and apoptosis of Tongue Squamous Cell Carcinoma-25(TSCC-25).Methods: Using quantitative Real-Time PCR(qRT-PCR)method for detecting TSCC-25 and normal tongue squamous cell(Normal Tongue Squamous Cell,NTSC)in the expression of microRNA-24.To construct the pGcsIL-GFP-IIliR-24 lentivirus expression vector and transform it into TSCC-25,then detect the expression difference of microRNA-24 in TSCC-25 was compared by the same operation method.The proliferation of TSCC-25 in transfection group,negative control group and blank control group was detected by MTT method,and the apoptosis of the three groups were detected by flow cytometry.Results : The microRNA-24 of NTSC expression was significantly higher than TSCC-25(P < 0.05).The amount of the expression of microRNA-24 that transfected group also significantly more often than not transfected group(P < 0.05).There was no significant difference between the negative control group and blank control group(P > 0.05).Using the method of MTT to detect cell proliferation rate,which was significantly decreased after transfected group(P < 0.05).Flow cytometry showed a significant increase in apoptosis of TSCC-25 after transfected with microRNA-24(P<0.05).Conclusion : Stable transfecting microRNA-24 into TSCC-25 can effectively inhibit the proliferation of TSCC-25,and this experiment can lay a foundation for further animal experiments and the study of microRNA-24 regulating target gene of TSCC-25.
