Nowadays,obesity has become a major cause of human death worldwide.Obesity,the reason of multiple chronic disease,can lead to ischemic heart disease,diabetes,fatty liver,cancer,atherosclerosis and degenerative diseases etc.Research on obesity has gained more attentions,but progress is limited.In this study,we found that TSP knockout(KO)mice are substantially resistant to high fat diet induced body weight gain.Mechanistically,we demonstrate that TSP KO mice exhibit similar amount of food intake,this is also evident by equal expression levels of neuropeptide in hypothalamus of the brain,which controls appetite.Moreover,these mice show similar intestinal absorption,and increased 02 consumption and C02 production,suggesting that TSP KO mice exhibit promoted basal metabolic level.Serum glucose level,adipose tissue,liver tissue and serum triglyceride level decreased significantly,hepatic glycogen level increased significantly.TSP knockout mice are more tolerant to exogenous glucose and insulin,as demonstrated by GTT and ITT assay.Levels of genes related to glycometabolism and lipid catabolism in TSP knockout mice are increased significantly,while those genes related to inflammation are decreased significantly.Our study reveals that TSP deficiency enhances the basal metabolic level in mice,promotes glycometabolism in liver and lipid catabolism in adipose tissue,eventually effectively resist obesity induced by high-fat diet.This discovery provides a novel therapeutic target for obesity treatment.
