Objective: 1.To investgate the rule of selecting acupoints by electro-acupuncture at single acupoint and compatible acupoints in the treatment of insomnia symptoms;2.To detect the changes of spatial learning and memory ability of sleep deprivation model rats before and after acupuncture by water maze test.3.To study the regulation mechanism of electro-acupuncture on sleep-related neurotransmitters in hippocampus of sleep deprivation model rats,and to study the therapeutic effect of electro-acupuncture at single and combined acupoints on insomnia,so as to provide theoretical basis for the neurobiological mechanism of insomnia and clinical acupuncture treatment of insomnia.Methods: A total of 60 healthy male SD rats were randomly divided into 6 groups(10rats in each group),namely the normal group,model group,baihui group,shenmen group,sanyin-jiao group and baihui-shenmen-sanyin-jiao compatibility group(" sancai group " for short).(1)Adaptive feeding was conducted one week before the start of the experiment,and a sleep deprivation model was established by intraperitoneal injection of phenylchlorophenylalanine(PCPA).The circadian rhythm of the rats disappeared and their daily activities were frequent,suggesting that the model was successful.(2)Morris water maze was used to evaluate the cognitive function of rats in each group for 4 consecutive days,and conduct positioning navigation and space exploration experiments.(3)building 2days after the successful treatment,treatment after 1 h,using Western Blot method(Western Blot)detection in the hippocampus of rats learning and memory related protein kinase A-C beta subunits(PKA-C?),cyclic adenosine monophosphate response element binding protein(P-CREB),brain derived neurotrophic factor(BDNF)and tropomyosin receptor kinase(TrkB)expression.Results:(1)the water maze navigation experiment showed that the incubation period of shenmen point group,baihui group,sanyinjiao group and compatibility group was significantly shortened(p < 0.05),and the incubation period of compatibility group waslower than that of the other three treatment groups(p < 0.05).Space exploration,the experimental results show that the model group rats through the platform the number of the original position significantly reduced(p < 0.05),the group,the three vaginal intercourse and acupuncture point group through increased significantly(p < 0.05),while the number of times the compatibility group was obviously more than the other three treatment group,suggest acupuncture single cavity god door,best can and compatibility were significantly improved the insomnia rats spatial working memory ability,among which improve compatibility effect is best.(2)rats compared with normal group,model group rats hippocampal tissues of BDNF,TrkB,P-after CREB and PKA-C beta protein expression significantly decreased(P < 0.01,P < 0.05,P < 0.05,P < 0.01),acupuncture after god door hole inside the hippocampus BDNF,TrkB and PKA-C beta expression quantity significantly increased(P < 0.05,P <0.05,P < 0.05),needle will point after TrkB and PKA-C beta expression significantly raised(P < 0.05,P < 0.05),Acupuncture after three vaginal intercourse acupuncture point,the expression of BDNF and TrkB volume significantly raised(p < 0.05,p < 0.05),after acupuncture acupoints compatibility of BDNF,TrkB,p-after CREB and PKA-C beta expression significantly raised(p < 0.01,p < 0.05,p < 0.01),and the expression of BDNF significantly higher than the group(p < 0.05),amount of PKA-C beta protein expression is significantly higher than vaginal intercourse three groups(p < 0.05),suggesting acupoints compatibility of insomnia rats hippocampal rise in improving the expression of protein involved in learning and memory function is better than that of single cavity.Conclusion: 1.Electroacupuncture can effectively regulate the learning and memory dysfunction caused by sleep deprivation in rats,both in terms of behavior and expression of learning and memory related proteins.2.Acusector can activate the hippocampal neurons in learning and memory function related to PKA-C beta-P-after CREB-BDNF/TrkB signal regulating circuit,the influence of different degree of the expression of the protein,and the effect of acupoints compatibility effect is superior to the single cavity,speculated that it will lead to a variety ofneural physiological changes and promote differentiation,regeneration and survival of hippocampal nerve cells,thereby improve insomnia rats learning and memory function.
