| Considerable attention is being paid to traditional Chinese medicine(TCM)because of its good pharmacodynamics and few side effects.However,there are some problems that have affected the development of TCM,such as unknown ingredients and unstable quality.Nowadays,fingerprint techniques are important for the quality control of TCM,as these methods can integrally reflect the complex composition of TCM.HSCCC is widely used in the separation and preparation of TCM.The high resolution and reproducibility of HPLC and HSCCC are similar,so HSCCC is suitable for fingerprint of TCM,as it can be considered equivalent to HPLC.Moreover,HSCCC analysis has advantages in samples pretreatment.Therefore,the HSCCC fingerprint was accurate,simple and fast,and can provide a new technical method for fingerprinting.Coumarin is one of the chemical constituents of many natural plants.It has strong physiological activities in many aspects,such as anti-inflammatory,anti-bacterial,anti-HIV and so on.Most coumarins have moderate polarity,which is suitable for the study of HSCCC.In this paper,the Chinese herbal medicine of Cnidium monnieri(L.)Cusson,Psorslea corylifolia L and Angelica dahurica Benth.et Hook,which are rich in coumarins,were selected for fingerprint study.Firstly,three extraction methods and different conditions of traditional Chinese medicine were optimized.Secondly,partition coefficient:K were determined through using TLC and HPLC methods,n-hexane-ethyl acetate-methanol-water was selected as the solvent system and the flow rate and injection volume were optimized.Finally,different batches of traditional Chinese medicines were carried out by HSCCC analysis.The HSCCC fingerprints of traditional three Chinese medicines were successfully established.In this study,coumarins were analyzed by high-speed countercurrent chromatography and fingerprints of three traditional Chinese medicines were established.It can be used for the quality control of Cnidium monnieri(L.)Cusson,Psorslea corylifolia L and Angelica dahurica Benth.et Hook.It also provides a new method for the fingerprint study of traditional Chinese medicine,which has important scientific significance.Firstly,HSCCC fingerprints can be used to identify Chinese medicines and their counterfeits easily,quality problems can be solved in the market of traditional Chinese medicine at the source.Secondly,the fingerprint of three TCM were established,which can be used for quality inspection in different producing areas and harvesting periods and provide a simple and effective method to solve the suboptimal problem in the market of traditional Chinese medicine.Finally,the HSCCC method can avoid the complex pretreatment of samples and can be used in the study of many batches of medicinal materials comparing with HPLC method.In this paper,HSCCC method is used for the first time to analyze the fingerprint of three kinds of TCM.This technique can be extended to the quality control of other TCM and can provide quality assurance for TCM on the international market. |
