Objective:To explore hypoglycemia mechanism of Rubus root polysaccharide on type 2 diabetic mice.Methods:Ten C57 male mice(18±2g)were randomly selected as normal control group The remaining 70 mice were fed high-fat and high-sugar diet combined with STZ intrapulmonary injected to construct the model of T2DM.Fasting blood glucoses greater than 7.8 mmol/L and was regarded as T2DM model.Fifty-eight successful T2DM mice were randomly divided into five groups accorded to their fasting blood glucose:model control group,metformin group(200 mg/kg),low-dose group(200 mg/kg),medium-dose group(400 mg/kg)and high-dose group(600 mg/kg).Normal control group and model group were given normal saline,and Rubus root polysaccharide groups were given different doses of Rubus root polysaccharide by gavage.The mice in each group were given Rubus root polysaccharide for 56 days(8 weeks).After 8 weeks,the levels of fasting blood glucose(FBG),triglyceride(TG),total cholesterol(TC)in the serum of mice in each group were detected.The liver of mice in each group was collected to determine AMP-activated protein kinase(AMPK),phosphoenolpyruvate cocarboxylase(PEPCK),glucose 6 phosphatase(G-6-P).Results:1.T2DM model establishment:1)After high-fat and high-sugar feeding for one week and intrapulmonary injection of STZ,there were 58 mice with fasting blood glucose level of?7.8mmol/L,and the rate of formation was 83%.2)Compared with the control group,the mice with fasting blood glucose?7.8mmol/L showed the phenomenon of "multiple drinks,more urine,and lower body weight".2.Hypoglycemic effect of Rubus root polysaccharide on T2DM mice:1)General state:Compared with the model group,the phenomenon of "drinking more food,eating more urine,lowering body weight" in each dose group of R.condenses polysaccharides was improved;2)Compared with the model group,the liver index of metformin(200 mg/kg)and high dose(rhodium saponin)(600 mg/kg)group was significantly lower(P<0.01),in the polysaccharide of Rubus The liver index of mice in the dose(400mg/kg)group was significantly lower(P<0.05).3)Compared with the control group,the serum levels of FBG and OGTT in the model group were significantly increased(P<0.01);compared with the model group,the metformin(200 mg/kg)group and the high dose of Rubus root polysaccharide(600 mg/kg)The levels of FBG and OGTT in the seram of the mice in the group were significantly decreased(P<0.01).The levels of FBG and OGTT in the serum of the mice in the dose of 400mg/kg were significantly lower(P<0.05);4)Compared with the control group,the levels of TC,TG and LDL in the serum of the model group were substantially increased(P<0.01).Compared with the model group,the metformin(200 mg/kg)group and the high dose of Rubus root polysaccharide(600 mg/The levels of TC,TG and LDL in the serum of mice in kg group were significantly decreased(P<0.01).The levels of TC,TG and LDL in the serum of mice in the dose of rhubarb root polysaccharide(400mg/kg)were significantly lower(P<0.05);5)Compared with the control group,the AMPK level in the liver tissue of the model group was significantly decreased(P<0.01);compared with the model group,the metformin(200 mg/kg)group and the high dose(600 mg/kg)group of R.condenses polysaccharides were small.The level of AMPK in the serum of rats was significantly increased(P<0.01);the level of AMPK in the liver tissue of mice in the dose of decoction root polysaccharide(400mg/kg)was significantly increased(P<0.05);6)Compared with the control group,levels of PEPCK and G-6-P in the liver tissue of the model group were significantly increased(P<0.01).Compared with the model group,the high dose(600 mg/kg)group and the metformin of the rhizosphere polysaccharides.Levels of PEPCK and G-6-P in the liver tissue of mice(200mg/kg)were significantly decreased(P<0.01).Conclusion:Rubus root polysaccharide could efectively lower blood glucose and plasma lipid,activate the AMPK signaling pathway and inhibited by two key enzymes PEPCK and G-6-P involved in the AMPK signaling pathway.
