Objectives:To explore the evaluation value of the LETM1 expression in colorectal carcinoma(CRC)in the prognosis of patients with CRC.To provide a reference for the prevention and cure of CRC through observing the relationship between the LETM1 and cancer stem cells markers.Methods:The western blotting was used to detect the expression of the LETM1,SOX9,CD44 and LGR5 in the two cell lines of the colon carcinoma HT29 and HCT116 when they were cultured to 70%fused.In addition the immunohistochemistry was used to detect the expression of the LETM1,SOX9,CD44 and LGR5 in the selected tissue samples of 102 patients with CRC and 43 patients with mucosa adjacent to CRC that have complete follow-up data as well as tissue microarray(The median follow-up time was 112 months).Besides,the correlation between the expressions of the LETM1 and the clinical-pathologic features of CRC,the LETM1 and the SOX9,CD44 and LGR5 were analyzed respectively;and the Kaplan-Meier survival analysis method was used to explore the relationship between the expression of the LETM1 and the prognosis of the CRC patients.Results:1.Positive expression of the LETM1,CD44,SOX9 and LGR5 have been observed in the colon carcinoma HT29 and HCT116 cells.2.The positive expression rate of the LETM1 in the CRC tissues(81.4%,83/102)was significantly higher than that in the adjacent mucosa tissues(44.2%,19/43)(p<0.05).3.The expression of the LETM1 was significantly correlated with TNM stage,lymph node metastasis and distant metastasis in patients with CRC(p<0.05).4.The overall survival and disease-free survival of CRC patients with positive expression of the LETM1 were significantly lower than those with negative expression ofthe LETMl(p<0.05).5.The LETM1 expression was an independent prognostic factor for the overall survival and disease-free survival of CRC patients.6.The LETM1 was positively correlated with cancer stem cells markers,the CD44(p=0.002),SOX9(p=0.002)and LGR5(p=0.018).Conclusion:1.The expression of the LETM1 is one of the risk factors affecting the prognosis of patients with CRC.2.The LETM1 may be a potential cancer stem cells marker.
