| Objective To Preliminarily Study on the apoptosis of Bakuchiol on hepatocellular carcinoma HepG-2 cells.Methods HepG-2 cells were used as experimental subjects.The anti-tumor activity of Bakuchiol on hepatocellular carcinoma HepG-2 cells was assessed by MTT assay.The effect of Bakuchiol on the morphology of HepG-2 cells was observed by HE staining.Cell cycle changes and apoptosis of hepatocellular carcinoma HepG-2 cells were detected by flow cytometry.The effect of Bakuchiol on DNA of hepatocellular carcinoma HepG-2 cells was observed by agarose gel electrophoresis.Results Compared with the control group,Bakuchiol significantly inhibited the proliferation of HepG-2 cells(P<0.01),and its inhibitory effect was positively correlated with time and concentration within 2.5-20 μM and 24-72 h.Compared with the control group,cell spacing increased,nucleus pyknosis,deep staining and cell membrane shrinkage were observed after administration of Bakuchiol in HepG-2 cells,which were typical apoptotic morphology.The degree of apoptosis increased with the time and the concentration of the drug administered.Bakuchiol induced apoptosis in HepG-2 cells(P<0.01),and its apoptosis rate was enhanced with the increase of time and concentrations within 2.5-20 μM and 24-48 h.Compared with the control group,the cell cycle distribution of hepatocellular carcinoma HepG-2 cells seemed to be affected after treated with increasing concentrations of Bakuchiol,presents as the proportion of cells in G0/G1 phase was significantly decreased(P<0.01),while the proportion of S phase was significantly increased(P<0.01).Compared with the control group,the DNA of hepatocellular carcinoma HepG-2 cells was cleaved after treated with Bakuchiol in different concentration for 48h,showing typical trapezoidal bands.Conclusion Bakuchiol can significantly induce the apoptosis of hepatocellular carcinoma HepG-2 cells,and its mechanism may be related to cell cycle arrest. |
