| Objective:1.The effects of ERK1/2,p-ERK1/2,CREB,p-CREB and BDNF protein expression in brain tissue of rats in each group were observed by acupuncture at the upper and middle points of CUMS rats.Related molecular mechanisms.2.To observe the abundance and diversity of intestinal microbes in the fecal DNA of acupuncture CUMS rats in Shangxing and Renzhong points,and to explore the intestinal tract of CUMS group,acupuncture acupuncture group and acupuncture non-acupoint group.Changes between the microbes.Methods:72 SPF male Sprague-Dawley rats were randomly divided into blank group,CUMS group,acupuncture acupoint group,acupuncture non-acupoint group,blank acupoint group and blank non-acupoint group,with 12 rats in each group.Except for the blank group,the blank acupoint group,and the blank non-acupoint group,the rest of the rats were prepared by the CUMS method for the depression rat model for 4 weeks.After 4 weeks,acupuncture treatment was performed on acupuncture points,acupuncture non-acupoint group,blank acupoint group and blank non-acupoint group for 3 weeks,and acupuncture point group and acupuncture non-acupoint group after acupuncture treatment.The model group rats continued to perform CUMS modeling.After the treatment is over,conduct a behavioral test.After the end of behavior,the rat hippocampus tissue was taken.The expression levels of BDNF,ERK1/2,p-ERK1/2,CREB and p-CREB proteins in hippocampus were detected by Western blotting.Analysis and comparison of changes in 16Sr RNA species richness in feces of each group of rats were performed by using 16Sr DNA sequencing analysis.Results:1.Compared with the normal group:the CUMS group’s body weight,open field test related indicators,elevated cross maze and other behavioral indicators were significantly smaller than the normal group(P<0.01);the normal acupoint group and the normal non-acupoint group were not statistically significant.Difference(P>0.05).Compared with the CUMS group,the behavioral indexes of the acupuncture points group were significantly changed(P<0.01).There was no significant change in behavioral indicators such as acupuncture in non-acupoint group(P>0.05).2.Compared with the normal group:there was no significant difference in ERK1/2 and CREB protein content in hippocampus of CUMS group(P>0.05),while BDNF,p-ERK1/2 and p-CREB levels were significantly lower,with statistical difference(P<0.01);there was no significant difference between the normal acupoint group and the normal non-acupoint group(P>0.05).Compared with CUMS group,there was no significant difference in ERK1/2 and CREB protein content in hippocampus of acupuncture points group(P>0.05),while BDNF,p-ERX1/2 and p-CREB were high in content.Statistical significance(P<0.01);the protein content in hippocampus of acupuncture group was not significantly changed(P>0.05).3.Compared with the normal group:the genus of the intestinal flora of the CUMS group was significantly different from the normal group(P<0.05)in the taxonomic level of the portal,genus,genus,genus and genus.Compared with the CUMS group,the abundance index of the acupuncture points group was significantly different from that of the CUMS group(P<0.01).There was no significant difference between the acupuncture point group and the CUMS group(P>0.05).).Conclusions:l.Acupuncture at ShangXing and RenZhong points has the effect of improving depression-like behavior in CUMS rats,which can reverse the behavioral differences of rats,and its mechanism may occur through BNDF-ERK-CREB signaling pathway.2.Acupuncture at ShangXing and RenZhong points of rats,it was observed that there were significant differences in the diversity and abundance of intestinal microbes between the intestinal microbes of the acupuncture points and the CUMS rats,Its mechanism of action may be related to the flora-intestinal-brain axis. |
