BackgroundRenal cell carcinoma is the third most common tumor in the urinary system following bladder and prostate cancer.The incidence has increased by years in recent years.Clear cell RCC(ccRCC)is the most common subtype of RCC,accounting for 80-90%.the clinical symptoms of patients with renal cancer are not obvious,and more often manifested as advanced symptoms when diagnosed,in recent years,about 25-30%of patients with initial diagnosis may be associated with tumor metastasis.At present,the main treatment methods for RCC rely on the surgical method,and the cancer is not sensitive to radiotherapy and chemotherapy.The recurrence and metastasis rate of postoperative patients is also high,Adjuvant therapy such as targeted therapy can be paid more and more attention as adjunctive therapy for patients with advanced cancer or postoperative.Although many biomarkers associated of renal cell carcinoma have been discovered,it is still necessary to explore highly specific biomarkers that affect the prognosis of renal cell carcinoma.Receptor-interacting protein(RIP)family kinases are an important classes of cellular stress sensors that are closely related to members of the interleukin-1 receptor-associated kinase(IRAK)family.To date,five RIPK family members have been discovered.The receptor-interacting serine/threonine protein kinase-2(RIPK2)is a kinase containing a caspase recruitment domain that is involved in cellular inflammation and immune processes.Recent studies have shown that RIPK2 activates downstream NF-?B signaling and expression of tumor-associated genes and is involved in the infiltration and metastasis of some cancers.The role of RIPK2 in renal cell carcinoma is not clear.Therefore,relevant research will help to further explain the correlation between RIPK2 and the development of renal cancer,and provide a new marker of renal cell cancer for the diagnosis and prognosis.ObjectiveTo determine the expression of RIPK2 in renal cell carcinoma and the correlation between RIPK2 with the clinicopathological features based on TCGA database,to explore the correlation between RIPK2 and prognosis of renal cell carcinoma.Methods1.Using HPA database to select differentially expressed genes in renal cancer,and analyze the protein expression and correlation with clinical prognosis.2.Using UALCAN database to analyze the relationship between RIPK2 gene expression level and clinical pathological parameters of ccRCC.3.Using GEPIA database and Oncolnc database to analyze the relationship between RIPK2 expression and prognosis.Results1.The HPA database shows that the expression of RIPK2 in ccRCC tissues is higher than that of normal tissues and is related to the prognosis of patients.2.UALCAN analysis:The expression of RIPK2 gene in ccRCC tissues was significantly higher than that in normal tissues(P=1.96E-12);However,this expression was not significantly correlated with the gender,age and ethnicity of patients,but correlated with tumor staging.,grading and molecular subtype.3.The PEPIA showed that the expression of RIPK2 gene was significantly increased in renal cell carcinoma(p<0.001),and the survival rate of disease-free of high RIPK2 gene expression was significantly decreased(P=0.012).Correspondingly,Oncolnc data analysis showed that the survival rate of tumor patients with high RIPK2 gene expression was significantly lower(P=2.36E-07).ConclusionOur study demonstrates that the expression of RIPK2 in ccRCC was significantly higher than that in normal tissue,and correlated with tumor stage,grade and molecular subtype.RIPK2 is associated with prognosis,and high expression of RIPK2 predicts poor prognosis in ccRCC.RIPK2 can be used as an independent prognostic factor for ccRCC.
