| Purpose In this study," spleen strengthening,blood nourishing and wind expelling method" was used to treat atopic dermatitis due to spleen deficiency and blood dryness.The curative effect of " spleen strengthening,blood nourishing and wind expelling method" on patients with atopic dermatitis due to spleen deficiency and blood dryness was preliminarily discussed by observing the changes of SCORAD score before and after taking the medicine and monitoring the expressions of serum IgE,IL17,IL18 and IL33 before and after taking the medicine.Taking hairless mice as experimental objects,the immunofluorescence staining of hairless mice treated with drugs with different drug concentrations of Jianpi Yangxue Qufeng Recipe and positive control drug loratadine were observed.The total optical density values of TAK1,TLR2,TLR4,P-P65 and MyD88 were determined by western blot experiment,and statistical analysis was carried out to preliminarily explore the effect of Jianpi Yangxue Qufeng Recipe on skin inflammation mechanism in animal experiments.Method1.Clinical observation Through clinical randomized controlled experiments,60 cases were included and randomly divided into treatment group(Jianpi Yangxue Qufeng Fang group)and control group(loratadine group),with 30 cases in each group.The course of treatment was 4 weeks,followed up for 2 months.Relevant data(detection of serum IgE,indicators of interleukin 17,18,33)were observed before treatment,during treatment(week 2)and after treatment(week 4)respectively and entered into Excel table.SPPS22 software was used for statistics and comparative analysis,and conclusions were drawn.2.Animal experimentsEighteen hairless mice(SKH1 hairless mice)without significant atopy were randomly divided into 6 groups by random control test,3 mice in each group,namely normal group,adhesive tape model group,positive control group,and high,medium and low dose groups according to different concentrations of Jianpi Yangxue Qufeng prescription.And according to the different groups,the normal group does not do the processing;The adhesive tape model group,positive control group,high,medium and low drug concentration groups were fed with normal saline,loratadine solution,high,medium and low drug concentration respectively,and back skin tissues were taken for immunofluorescence staining,and the staining conditions of each group were compared.After half of the fresh materials were frozen at-80,the subsequent western blot experiment was carried out,and the observation data(total optical density values of TAK1,TLR2,TLR4,P-P65 and MyD88)were recorded and entered into Excel table,and the SPPS 22 software was used for statistics and comparative analysis to draw a conclusion.Result1.Clinical observation results1.1 the total effective rate after 2 weeks of treatment with Jianpi yangxue qufeng recipe is 72.34%,and the total effective rate after 4 weeks of treatment with Jianpi yangxue qufeng recipe is 93.33%.there is a significant difference between the two groups(P<0.05).1.2 follow-up results: the relapse rate of Jianpi yangxue qufeng recipe was 11.53%,and that of loratadine group was 28.57%,P<0.05,with statistical significance.1.3 Adverse Events: No adverse events occurred in the two groups,with good safety.2.Animal experiment results2.1 compared with the normal control group,the fluorescence intensity of Myd88,NF-κB,TLR2,TLR4 in the model group increased,and the expression of corresponding proteins increased.After drug treatment,the fluorescence intensity of Myd88,NF-κB,TLR2,TLR4 decreased,and the expression of corresponding proteins decreased in a dose-dependent manner.2.2 the expression of TLR2,TLR4,My D88,NF-κB protein in skin lesions of the spleen invigorating,blood nourishing and wind expelling prescription group was significantly higher than that of the blank group(**p<0.01).Conclusion " Jianpi Yangxue Qufeng Method" has certain curative effect in treating AD.Compared with the oral western medicine group,the symptoms of the patients are obviously improved after 4 weeks of treatment,and no obvious adverse reactions are found in the treatment.In the long run,this method has certain advantages in treating atopic dermatitis with spleen deficiency and blood dryness syndrome,which is worthy of clinical promotion.The results of this study show that Tight Junction Protein is closely related to the pathogenesis of AD.Serum IgE,IL-17,IL-18,IL-33 can be used as important indicators in the occurrence and development of AD diseases. |
