Objective1.To investigate the expression of DEUP1 in hepatocellular carcinoma and its clinical significance.2.To investigate the relationship between the methylation status of DEUP1 promoter in hepatocellular carcinoma and its clinicopathological characteristics,and to analyze the relationship between the methylation status of DEUP1 promoter and protein expression.Methods1.Collecting 60 cases of hepatocellular carcinoma and matched paracancerous tissue specimens from the first hospital of Zhengzhou University from January 2016 to December 2016 with surgical resection and pathological diagnosis.2.Real-time quantitative PCR and western blot were used to detect the expression level of DEUP1 in 60 cases of hepatocellular carcinoma and matched paracancerous tissues.The relationship between the expression of DEUP1 and clinicopathological features was analyzed.The database was used to predict the relationship between DEUP1 mRNA expression levels and overall patient survival.3.The methylation status of DEUP1 promoter in hepatocellular carcinoma and matched paracancerous tissues was predicted by using methylation database,and the methylation status of DEUP1 promoter in 60 hepatocellular carcinoma and matched paracancerous tissues was detected by bisulfite sequencing(BSP),and its relationship with clinicopathological characteristics was analyzed.4.Statistical analysis of the correlation between the degree of methylation of DEUP1 promoter and its protein expression.Resoults1.The relative expression levels of DEUP1 mRNA and protein in 60 cases of hepatocellular carcinoma were 0.00152±0.00036?0.054±0.026,and the relative expressions of DEUP1 mRNA and protein in adjacent tissues were 0.00273±0.0095?0.266±0.088.Statistical analysis showed that The expression of DEUP1 mRNA and protein in hepatocellular carcinoma tissues was significantly lower than that in matched paracancerous tissues.2.Correlation analysis between the expression of DEUP1 protein in hepatocellular carcinoma tissues and clinicopathological features of patients showed that the expression level of DEUP1 protein was related to TNM stage and tumor differentiation.Kaplan-Meier Plotter analysis showed that the overall survival rate of patients with high expression of DEUP1 mRNA was higher than that of patients with low expression.3.The methylation ratio of DEUP1 promoter in 60 cases of hepatocellular carcinoma tissues was(76.6±13.9)%,and the methylation ratio of DEUP1 promoter in adjacent tissues was(15.9±10.0)%.Statistical analysis showed that the methylation level of DEUP1 promoter in hepatocellular carcinoma tissues was significantly higher than that in paired adjacent tissues.Correlation analysis between the methylation status of the DEUP1 promoter and the clinicopathological features of the patient indicates that the methylation status of the DEUP1 promoter is related to the TNM stage and the degree of tumor differentiation.4.Correlation analysis of DEUP1 promoter methylation and protein expression in hepatocellular carcinoma showed that the degree of methylation of DEUP1 promoter was negatively correlated with its protein expression level.Conclusion1.DEUP1 expression in hepatocellular carcinoma tissues was significantly lower than that in matched paracancerous tissues.2.The expression of DEUP1 in hepatocarcinoma tissues is related to the TNM stage and tumor differentiation of liver cancer patients.The overall survival rate of patients with high expression of DEUP1 mRNA is higher than that of patients with low expression.3.The degree of methylation of DEUP1 promoter in hepatocellular carcinoma tissues was significantly higher than that in matched paracancerous tissues.The degree of methylation was related to the TNM stage and tumor differentiation of liver cancer patients.4.The degree of methylation of DEUP1 promoter in hepatocellular carcinoma tissues was negatively correlated with its protein expression level.
